In the vertebrate inner ear, the ability to detect angular head movements lies in the three semicircular canals and their sensory tissues, the cristae. The molecular mechanisms underlying the formation of the three canals are largely unknown. Malformations of this vestibular apparatus found in zebrafish and mice usually involve both canals and cristae. Although there are examples of mutants with only defective canals, few mutants have normal canals without some prior sensory tissue specification, suggesting that the sensory tissues, cristae, might induce the formation of their non-sensory components, the semicircular canals. We fate-mapped the vertical canal pouch in chicken that gives rise to the anterior and posterior canals, using a fluorescent, lipophilic dye (DiI), and identified a canal genesis zone adjacent to each prospective crista that corresponds to the Bone morphogenetic protein 2 (Bmp2)-positive domain in the canal pouch. Using retroviruses or beads to increase Fibroblast Growth Factors (FGFs) for gain-of-function and beads soaked with the FGF inhibitor SU5402 for loss-of-function experiments, we show that FGFs in the crista promote canal development by upregulating Bmp2. We postulate that FGFs in the cristae induce a canal genesis zone by inducing/upregulating Bmp2 expression. Ectopic FGF treatments convert some of the cells in the canal pouch from the prospective common crus to a canal-like fate. Thus, we provide the first molecular evidence whereby sensory organs direct the development of the associated non-sensory components, the semicircular canals, in vertebrate inner ears.

In vertebrati orecchio interno, la capacit&agrave; di rilevare movimenti angolari del capo si trova in tre canali semicircolari e loro tessuti sensoriale, i cristae. I meccanismi molecolari che sottendono la formazione dei tre canali sono in gran parte sconosciuti. Malformazioni di questo apparato vestibolare trovano nello zebrafish e topi di solito comportano entrambi i canali e cristae. Scleraxis (Scx) is a basic helix-loop-helix transcription factor expressed in tendon and ligament progenitor cells and the differentiated cells within these connective tissues in the axial and appendicular skeleton. Unexpectedly, we found expression of the Scx transgenic reporter mouse, Scx-GFP, in interdental cells, sensory hair cells, and cochlear supporting cells at embryonic dayÂ 18.5 (E18.5). We evaluated Scx-null mice to gain insight into the function of Scx in the inner ear. Paradoxical hearing loss was detected in Scx-nulls, with ~50% of the mutants presenting elevated auditory thresholds. However, Scx-null mice have no obvious, gross alterations in cochlear morphology or cellular patterning. Moreover, we show that the elevated auditory thresholds correlate with middle ear infection. Laser interferometric measurement of sound-induced malleal movements in the infected Scx-nulls demonstrates increased impedance of the middle ear that accounts for the hearing loss observed. The vertebrate middle ear transmits vibrations of the tympanic membrane to the cochlea. The tensor tympani and stapedius muscles insert into the malleus and stapes via distinct tendons and mediate the middle ear muscle reflex that in part protects the inner ear from noise-induced damage. Nothing, however, is known about the development and function of these tendons. Scx is expressed in tendon progenitors at E14.5 and differentiated tenocytes of the stapedius and tensor tympani tendons at E16.5-18.5. Scx-nulls have dramatically shorter stapedius and tensor tympani tendons with altered extracellular matrix consistent with abnormal differentiation in which condensed tendon progenitors are inefficiently incorporated into the elongating tendons. Scx-GFP is the first transgenic reporter that identifies middle ear tendon lineages from the time of their formation through complete tendon maturation. Scx-null is the first genetically defined mouse model for abnormal middle ear tendon differentiation. Scx mouse models will facilitate studies of tendon and muscle formation and function in the middle ear.

Scleraxis is required for differentiation of the stapedius and tensor tympani tendons of the middle ear.

Scleraxis&#xB294; &#xC911;&#xAC04; &#xADC0; stapedius &#xBC0F; &#xD150;&#xC11C; tympani &#xD798; &#xC904;&#xC758; &#xBD84;&#xD654;&#xC5D0; &#xB300; &#xD55C; &#xD544;&#xC694; &#xD569;&#xB2C8;&#xB2E4;.

Scleraxis (Scx) es un factor de transcripci&oacute;n de h&eacute;lice-bucle-h&eacute;lice b&aacute;sica expresado en c&eacute;lulas progenitoras de tendones y ligamentos y las c&eacute;lulas diferenciadas dentro de estos tejidos conectivos en el esqueleto axial y apendicular. Inesperadamente, encontramos la expresi&oacute;n del rat&oacute;n transg&eacute;nico reportero Scx, Scx-GFP, en c&eacute;lulas interdentales, sensoriales de las c&eacute;lulas ciliadas y c&eacute;lulas de soporte cocleares en el d&iacute;a embrionario 18,5 (E18.5). Evaluamos ratones Scx-null para profundizar en la funci&oacute;n de Scx en el o&iacute;do interno. Hipoacusia parad&oacute;jico fue detectada en Scx-nulos, con ~ 50% de los mutantes que presenta elevados umbrales auditivos. Sin embargo, Scx-null ratones tienen ningunas alteraciones evidentes, brutas en morfolog&iacute;a coclear o patrones celulares. Por otra parte, demostramos que los umbrales auditivos elevados se correlacionan con la infecci&oacute;n del o&iacute;do medio. Medici&oacute;n por l&aacute;ser interferom&eacute;trico de movimientos malleal inducida por el sonido en los infectados Scx-nulls demuestra mayor impedancia del o&iacute;do medio que representa la p&eacute;rdida de la audici&oacute;n observada. El o&iacute;do medio vertebrado transmite las vibraciones de la membrana del t&iacute;mpano a la c&oacute;clea. Los m&uacute;sculos tensor del t&iacute;mpano y stapedius Inserte el martillo y del estribo mediante distintos tendones y median el reflejo muscular del o&iacute;do medio que protege en parte el o&iacute;do interno del da&ntilde;o provocado por el ruido. Sin embargo, nada se sabe sobre el desarrollo y la funci&oacute;n de estos tendones. SCX se expresa en los progenitores de tend&oacute;n en E14.5 y distinguido tenocytes de tendones stapedius y tensor del t&iacute;mpano en E16.5-18.5. SCX-nulos tienen stapedius dram&aacute;ticamente menor y tensor del t&iacute;mpano tendones con la matriz extracelular alterada consistente con diferenciaci&oacute;n anormal en el cual progenitores tend&oacute;n condensada ineficiente se incorporan los tendones se alarga. SCX-GFP es el primer reportero transg&eacute;nico que identifica linajes de tend&oacute;n del o&iacute;do medio desde el momento de su formaci&oacute;n a trav&eacute;s de la maduraci&oacute;n completa del tend&oacute;n. SCX-null es el primero gen&eacute;ticamente definido modelo de rat&oacute;n para la diferenciaci&oacute;n de tend&oacute;n anormal de o&iacute;do medio. Modelos de rat&oacute;n SCX facilitar&aacute; estudios de tend&oacute;n y la formaci&oacute;n de los m&uacute;sculos y la funci&oacute;n en el o&iacute;do medio.

Scleraxis se requiere para la diferenciaci&oacute;n de los tendones del stapedius y tensor del t&iacute;mpano del o&iacute;do medio.

Cochlear inner hair cells (IHCs) use Ca(2+)-dependent exocytosis of glutamate to signal sound information. Otoferlin (Otof), a C(2) domain protein essential for IHC exocytosis and hearing, may serve as a Ca(2+) sensor in vesicle fusion in IHCs that seem to lack the classical neuronal Ca(2+) sensors synaptotagmin 1 (Syt1) and Syt2. Support for the Ca(2+) sensor of fusion hypothesis for otoferlin function comes from biochemical experiments, but additional roles in late exocytosis upstream of fusion have been indicated by physiological studies. Here, we tested the functional equivalence of otoferlin and Syt1 in three neurosecretory model systems: auditory IHCs, adrenal chromaffin cells, and hippocampal neurons. Long-term and short-term ectopic expression of Syt1 in IHCs of Otof (-/-) mice by viral gene transfer in the embryonic inner ear and organotypic culture failed to rescue their Ca(2+) influx-triggered exocytosis. Conversely, virally mediated overexpression of otoferlin did not restore phasic exocytosis in Syt1-deficient chromaffin cells or neurons but enhanced asynchronous release in the latter. We further tested exocytosis in Otof (-/-) hippocampal neurons and in Syt1(-/-) IHCs but found no deficits in vesicle fusion. Expression analysis of different synaptotagmin isoforms indicated that Syt1 and Syt2 are absent from mature IHCs. Our data argue against a simple functional equivalence of the two C(2) domain proteins in exocytosis of IHC ribbon synapses, chromaffin cells, and hippocampal synapses.
