&#x661F;&#x5F62;&#x80F6;&#x8D28;&#x7EC6;&#x80DE;&#x63A7;&#x5236;&#x4EA7;&#x540E;&#x5927;&#x8111;&#x4E2D;&#x901A;&#x8FC7; VEGF &#x4FE1;&#x53F7;&#x8F6C;&#x5BFC;&#x4E2D;&#x7684;&#x8FC1;&#x79FB;&#x4FC3;&#x8FDB;&#x8840;&#x7BA1;&#x652F;&#x67B6;&#x7684;&#x53D1;&#x5C55;&#x3002;

New neurons are constantly being generated in the postnatal subventricular zone. They have to migrate long distances via the rostral migratory stream (RMS) to reach their final destination in the olfactory bulb (OB). In adults, these neuronal precursors migrate in chains, ensheathed by astrocytic processes, and travel toward the OB along blood vessels (BVs) that topographically outline the RMS. The molecular and cellular mechanisms leading to the development of the RMS and the formation of the migration-promoting vasculature scaffold in the adult mice remain unclear. We now reveal that astrocytes orchestrate the formation and structural reorganization of the vasculature scaffold in the RMS and, during early developmental stages, the RMS contains only a few BVs oriented randomly with respect to the migrating neuroblasts. The first parallel BVs appeared at the outer border of the RMS, where vascular endothelial growth factor (VEGF)-expressing astrocytes are located. Gain-of-function and loss-of-function experiments revealed that astrocyte-derived VEGF plays a crucial role in the formation and growth of new BVs. Real-time videoimaging also showed that the migration of neuronal precursors in the developing RMS differs substantially from neuronal displacement in the adult migratory stream partially because of not yet fully developed vasculature scaffold. The downregulation of VEGF in vivo, specifically in the astrocytes of the developing RMS, affected the development of the vasculature scaffold and led to alterations in neuroblast migration. Altogether, our results demonstrate that astrocytes orchestrate the formation and growth of parallel BVs, crucial migration-promoting scaffolds in the adult migratory stream, via VEGF signaling.

Ils doivent migrer de longues distances via le flux migratoire rostral (RMS) d&#39;atteindre leur destination finale dans le bulbe olfactif (OB). St&auml;ndig werden neue Neuronen in der postnatalen subventrikul&auml;re Zone erzeugt. Wir zeigen nun, dass die Astrozyten zu orchestrieren, die Bildung und die Umstrukturierung der Gef&auml;&szlig;e Ger&uuml;st in den RMS und w&auml;hrend der fr&uuml;hen Entwicklungsstadien der RMS enth&auml;lt nur wenige BVs in Bezug auf die Migration von Neuroblasten zuf&auml;llig orientierte. Die erste parallele BVs erschien am &auml;u&szlig;eren Rand des RMS, wo vascular endothelial Wachstum factor (VEGF)-Ausdruck Astrozyten befinden. Gain of Function und Verlust der Funktion Experimente ergaben, dass die Gliazelle abgeleitete VEGF spielt eine entscheidende Rolle bei der Bildung und Wachstum von neuen BVs. Das Medikament VEGF in-vivo, speziell in den Astrozyten des entwickelnden RMS, beeinflusst die Entwicklung der Gef&auml;&szlig;e Ger&uuml;st und f&uuml;hrte zu Ver&auml;nderungen in Blast (Biologie)-Migration. Insgesamt unsere Ergebnisse zeigen, dass die Astrozyten der Entstehung und dem Wachstum der parallelen BVs, entscheidende Migration-F&ouml;rderung von Ger&uuml;sten im Erwachsenen wandernden Stream, orchestrieren &uuml;ber VEGF signalisieren.

Nuovi neuroni vengono costantemente generati nella zona subventricolare postnatale. Nuevas neuronas constantemente se est&aacute;n generando en la zona subventricular postnatal. Que tengan que emigrar largas distancias a trav&eacute;s de la corriente migratoria rostral (RMS) para llegar a su destino final en el bulbo olfatorio (OB). En los adultos, estos precursores neuronales migran en cadenas, rodeadas por los procesos astroc&iacute;ticos y viajan hacia el OB a lo largo de los vasos sangu&iacute;neos (BVs) que describen topogr&aacute;ficamente el RMS. Los mecanismos moleculares y celulares hacia el desarrollo de la RMS y la formaci&oacute;n del andamio vasculatura promoci&oacute;n de la migraci&oacute;n en los ratones adultos todav&iacute;a no est&aacute;n claros. Ahora nos revelan que astrocitos orquestaran la formaci&oacute;n y la reorganizaci&oacute;n estructural del andamio vasculatura en la RMS y, durante las primeras etapas del desarrollo, el RMS contiene s&oacute;lo unos BVs orientadas al azar con respecto a las migraci&oacute;n neuroblastos. Las BVs paralelos primeros aparecieron en la frontera exterior de la RMS, donde factor de crecimiento endotelial vascular (VEGF)-astrocitos expresando se encuentran. Ganancia de funci&oacute;n y de la p&eacute;rdida-de-funci&oacute;n experimentos revelaron que derivado del Astrocito VEGF desempe&ntilde;a un papel crucial en la formaci&oacute;n y crecimiento de nuevo BVs. videoimaging en tiempo real tambi&eacute;n demostr&oacute; que la migraci&oacute;n de precursores neuronales en la RMS en desarrollo difiere sustancialmente de desplazamiento neuronal en la corriente migratoria adulta parcialmente debido de no todav&iacute;a completamente desarrollado el andamio de la vasculatura. La regulaci&oacute;n a la baja de VEGF en vivo, espec&iacute;ficamente en los astrocitos de la RMS en desarrollo, afect&oacute; el desarrollo del andamio vasculatura y conducido a alteraciones en la migraci&oacute;n neurobl&aacute;stica. En conjunto, nuestros resultados demuestran que los astrocitos orquestaran la formaci&oacute;n y crecimiento de BVs paralelos, cruciales andamios de promoci&oacute;n de la migraci&oacute;n en el flujo migratorio adultos, v&iacute;a de se&ntilde;alizaci&oacute;n de VEGF.
