&#x611F;&#x5B98;&#x7684;&#x4EE3;&#x8868;&#x6027;&#x548C;&#x8611;&#x83C7;&#x8EAB;&#x4F53;&#x5916;&#x5728;&#x53CD;&#x9988;&#x795E;&#x7ECF;&#x5143;&#x7684; protocerebral &#x9053;&#x4E0E;&#x5B66;&#x4E60;&#x6709;&#x5173;&#x7684;&#x53EF;&#x5851;&#x6027;&#x3002;

Gamma-aminobutyric acid immunoreactive feedback neurons of the protocerebral tract are a major component of the honeybee mushroom body. They have been shown to be subject to learning-related plasticity and provide putative inhibitory input to Kenyon cells and the pedunculus extrinsic neuron, PE1. We hypothesize, that learning-related modulation in these neurons is mediated by varying the amount of inhibition provided by feedback neurons. We performed Ca(2+) imaging recordings of populations of neurons of the protocerebral-calycal tract (PCT) while the bees were conditioned in an appetitive olfactory paradigm and their behavioral responses were quantified using electromyographic recordings from M17, the muscle which controls the proboscis extension response. The results corroborate findings from electrophysiological studies showing that PCT neurons respond to sucrose and odor stimuli. The odor responses are concentration dependent. Odor and sucrose responses are modulated by repeated stimulus presentations. Furthermore, animals that learned to associate an odor with sucrose reward responded to the repeated presentations of the rewarded odor with less depression than they did to an unrewarded and a control odor.

Sie haben nachweislich unterliegen im Zusammenhang mit lernen Plastizit&auml;t und Bereitstellung von vermeintlichen hemmenden Input Kenyon Zellen und den Pedunculus extrinsischen Neuron, PE1. Wir vermuten, die lernen-bezogene Modulation in diese Neuronen vermittelt wird, indem man die Menge der Hemmung von R&uuml;ckmeldungen Neuronen bereitgestellt. The lateral line system of larval zebrafish is emerging as a model to study a range of topics in neurobiology, from hair cell regeneration to sensory processing. However, despite numerous studies detailing the patterning and development of lateral line neuromasts, little is known about the organization of their connections to afferent neurons and targets in the hindbrain. We found that as fish grow and neuromasts proliferate over the body surface, the number of afferent neurons increases linearly. The number of afferents innervating certain neuromasts increases over time, while it decreases for other neuromasts. The ratio of afferent neurons to neuromasts differs between the anterior and posterior lateral line system, suggesting potential differences in sensitivity threshold or spatial resolution. A single afferent neuron routinely contacts a group of neuromasts, suggesting that different afferent neurons can convey information about receptive fields along the body. When afferent projections are traced into the hindbrain, where a distinct somatotopy has been previously described, we find thatthis general organization is absent at the Mauthner cell. We speculate that directional input from the lateral line is less important at an early age, whereas the speed of the escape response is paramount, and that directional responses arise later in development. By quantifying morphological connections in the lateral line system, this study provides a detailed foundation to understand how hydrodynamic information is processed and ultimately translated into appropriate motor behaviors. Honeybees learn to associate an odor with sucrose reward under conditions that allow the monitoring of neural activity by imaging Ca(2+) transients in morphologically identified neurons. Here we report such recordings from mushroom body extrinsic neurons - which belong to a recurrent tract connecting the output of the mushroom body with its input, potentially providing inhibitory feedback - and other extrinsic neurons. The neurons' responses to the learned odor and two novel control odors were measured 24 h after learning. We found that calcium responses to the learned odor and an odor that was strongly generalized with it were enhanced compared with responses to a weakly generalized control. Thus, the physiological responses measured in these extrinsic neurons accurately reflect what is observed in behavior. We conclude that the recorded recurrent neurons feed information back to the mushroom body about the features of learned odor stimuli. Other extrinsic neurons may signal information about learned odors to different brain regions.

Fishes rely on the neuromasts of their lateral line system to detect water flow during behaviors such as predator avoidance and prey localization. While the pattern of neuromast development has been a topic of detailed research, we still do not understand the functional consequences of its organization. Previous work has demonstrated somatotopy in the posterior lateral line, whereby afferent neurons that contact more caudal neuromasts project more dorsally in the hindbrain than those that contact more rostral neuromasts (Gompel et al., 2001). We performed patch clamp recordings of afferent neurons that contact neuromasts in the posterior lateral line of anesthetized, transgenic larval zebrafish (Danio rerio) to show that larger cells are born earlier, have a lower input resistance, a lower spontaneous firing rate and tend to contact multiple neuromasts located closer to the tail than smaller neurons, which are born later, have a higher input resistance, a higher spontaneous firing rate and tend to contact single neuromasts. We suggest that early-born neurons are poised to detect large stimuli during the initial stages of development. Later-born neurons are more easily driven to fire and thus likely to be more sensitive to local, weaker flows. Afferent projections onto identified glutamatergic regions in the hindbrain lead us to hypothesize a novel mechanism for lateral line somatotopy. We show that afferent fibers associated with tail neuromasts respond to stronger stimuli and are wired to dorsal hindbrain regions associated with Mauthner-mediated escape responses and fast, avoidance swimming. The ability to process flow stimuli by circumventing higher order brain centers would ease the task of processing where speed is of critical importance. Our work lays the groundwork to understand how the lateral line translates flow stimuli into appropriate behaviors at the single cell level.

Les travaux pr&eacute;c&eacute;dents a d&eacute;montr&eacute; somatotopie dans la post&eacute;rieure de la ligne lat&eacute;rale, par lequel les neurones aff&eacute;rents qui contacter plus caudal neuromasts du projet plus sur le dos dans le cerveau post&eacute;rieur que celles que le contact plus rostral neuromasts (Gompel et coll., 2001). Nous avons effectu&eacute; patch clamp enregistrements des neurones aff&eacute;rents que contact neuromasts dans la ligne lat&eacute;rale post&eacute;rieure des transg&eacute;nique, anesth&eacute;si&eacute;, larve z&egrave;bre (Danio rerio) pour montrer que les plus grosses cellules naissent plus t&ocirc;t, ont une faible r&eacute;sistance d&#39;entr&eacute;e, une d&eacute;charge spontan&eacute;e plus faible note et ont tendance &agrave; communiquer avec multiples neuromasts situ&eacute; plus pr&egrave;s de la queue de petits neurones, qui sont n&eacute;s plus tard, ont une plus grande r&eacute;sistance d&#39;entr&eacute;e, une d&eacute;charge spontan&eacute;e sup&eacute;rieure note et ont tendance &agrave; communiquer avec neuromasts unique. Nous sugg&eacute;rons que les neurones pr&eacute;coces sont pr&ecirc;ts pour d&eacute;tecter des stimuli grands durant les premi&egrave;res &eacute;tapes du d&eacute;veloppement. La capacit&eacute; de traiter des stimuli de flux en contournant les centres du cerveau commande sup&eacute;rieurs faciliterait la t&acirc;che de traitement o&ugrave; la vitesse est d&#39;une importance cruciale. Notre travail jette les bases pour comprendre comment la ligne lat&eacute;rale se traduit par des stimuli de flux dans des comportements appropri&eacute;s au niveau de la cellule unique.

Fische verlassen sich auf die Neuromasts ihres Systems Seitenlinienorgan erkennen Wasserstrom w&auml;hrend Verhaltensweisen z. B. R&auml;uber-Vermeidung und Lokalisierung zum Opfer. W&auml;hrend das Muster der Neuromast Entwicklung ein Thema ausf&uuml;hrliche Forschung gewesen ist, verstehen wir die funktionellen Konsequenzen seiner Organisation noch nicht. Bisherige Arbeit zeigte Somatotopy in die hintere Seitenlinie, wobei afferente Neuronen, die mehr kaudale Neuromasts Kontakt mehr dorsal in das Hinterhirn als Projekt, die mehr K&ouml;rperabschnitt Neuromasts (Gompel Et Al., 2001) zu kontaktieren. Wir spielten Patch Clamp Aufnahmen von afferenten Neuronen Kontakt Neuromasts in die hintere Seitenlinie der &auml;sthetisierten, transgene larvalen Zebrab&auml;rbling (Danio Rerio) zeigen, dass gr&ouml;&szlig;ere Zellen fr&uuml;her geboren werden haben einen niedrigeren Eingangswiderstand, eine niedrigere spontane Ausl&ouml;sung bewerten und neigen dazu, mehrere Neuromasts liegt n&auml;her an den Schwanz, als kleinere Neuronen, die sp&auml;ter geboren werden, einen h&ouml;heren Eingangswiderstand haben kontaktieren, eine h&ouml;here spontane Ausl&ouml;sung bewerten und neigen dazu, die einzelnen Neuromasts zu kontaktieren. Wir schlagen vor, dass fr&uuml;h geborene Neuronen bereit sind, um gro&szlig;e Reize in der Anfangsphase der Entwicklung zu erkennen. Sp&auml;ter geborene Neuronen sind leichter angetriebene Feuer und somit empfindlicher auf lokalen, schw&auml;chere Str&ouml;me. Afferente Projektionen auf identifizierten Glutamat Regionen in das Hinterhirn veranlassen uns, einen neuen Mechanismus f&uuml;r Seitenlinienorgan Somatotopy vermuten. Wir zeigen, dass die afferente Fasern zugeordnete Tail Neuromasts auf st&auml;rkere Reize zu reagieren und sind so verschaltet, dorsal Hinterhirn Regionen zugeordnet Escape Mauthner-vermittelte Reaktionen und schnell, die Vermeidung schwimmen. Die M&ouml;glichkeit zum Fluss Reize verarbeiten unter Umgehung h&ouml;here Ordnung Gehirnzentren w&uuml;rde die Aufgabe der Verarbeitung erleichtern, wo Geschwindigkeit von entscheidender Bedeutung ist. Unsere Arbeit legt den Grundstein, um zu verstehen, wie die Seitenlinie Str&ouml;mung Reize in entsprechenden Verhaltensweisen auf der Ebene der einzelnen Zelle &uuml;bersetzt.

Abbiamo eseguito il patch clamp registrazioni dei neuroni afferenti che neuromasts contatti in linea laterale posteriore di anestetizzati, transgenici zebrafish larvale (Danio rerio) per dimostrare che le cellule pi&ugrave; grandi sono nati in precedenza, hanno una bassa resistenza d&#39;ingresso, un infornamento spontaneo inferiore tasso e tendono a contatto pi&ugrave; neuromasts che si trova pi&ugrave; vicino alla coda di neuroni pi&ugrave; piccoli, che sono nati pi&ugrave; tardi, hanno una pi&ugrave; alta resistenza di ingresso, un infornamento spontaneo pi&ugrave; elevato tasso e tendono a contatto singolo neuromasts. Suggeriamo che neuroni precoce-NATO sono pronti a rilevare grandi stimoli durante le fasi iniziali di sviluppo. Pi&ugrave; tardi nato neuroni sono pi&ugrave; facilmente spinti a fuoco e quindi suscettibili di essere pi&ugrave; sensibili ai flussi locali, pi&ugrave; deboli. Le proiezioni afferenti su regioni individuate glutamatergico il hindbrain ci portano a ipotizzare un nuovo meccanismo per linea laterale somatotopy. Mostriamo che fibre afferenti associati con coda neuromasts rispondono agli stimoli pi&ugrave; forti e sono cablati a regioni dorsali hindbrain associato a risposte di fuga Mauthner-mediata e veloce, evitare il nuoto. La capacit&agrave; di elaborare stimoli flusso di eludere i centri superiori del cervello di ordine dovrebbe facilitare il compito di elaborazione dove la velocit&agrave; &egrave; di importanza critica. Il nostro lavoro pone le basi per comprendere come la linea laterale si traduce stimoli di flusso in appropriati comportamenti a livello di singola cellula.

Peces dependen de la neuromasts de su sistema de l&iacute;nea lateral para detectar el flujo de agua en comportamientos tales como la evitaci&oacute;n de depredador y presa de localizaci&oacute;n. Mientras que el patr&oacute;n de desarrollo de neuromast ha sido un tema de investigaci&oacute;n detallada, que todav&iacute;a no entendemos las consecuencias funcionales de su organizaci&oacute;n. El trabajo previo ha demostrado somatotopy en la l&iacute;nea lateral posterior, por el que las neuronas aferentes que Contacta con m&aacute;s caudal neuromasts proyecto m&aacute;s dorsalmente en el cerebelo que aquellas que en contacto con neuromasts m&aacute;s rostral (Gompel et al., 2001). Nuestro trabajo sienta las bases para entender c&oacute;mo la l&iacute;nea lateral se traduce est&iacute;mulos de flujo en comportamientos apropiados a nivel unicelular.
