Tejido adiposo marr&oacute;n (BAT) se disipa energ&iacute;a qu&iacute;mica y genera calor para proteger a los animales del fr&iacute;o y la obesidad. Roedores poseen dos tipos de UCP-1 positivos adipocitos marrones derivadas de distintos linajes del desarrollo: adipocitos marrones &quot;cl&aacute;sicas&quot; se desarrollan durante la etapa prenatal, mientras que las c&eacute;lulas &quot;beige&quot; o &quot;brite&quot; que residen en el tejido adiposo blanco (WAT) desarrollan durante la etapa postnatal en respuesta al fr&iacute;o cr&oacute;nica o agonistas PPARy. Caracter&iacute;sticas inducible beige c&eacute;lulas hacen una diana terap&eacute;utica prometedora para el tratamiento de la obesidad, sin embargo, se desconoce la relevancia de este tipo celular en los seres humanos. En el presente estudio, determinamos las firmas de gene que cl&aacute;sico adipocitos marrones y beige c&eacute;lulas inducidas por un espec&iacute;fico PPARy agonista rosiglitazona en ratones. Posteriormente aplicamos los datos transcripcionales a los seres humanos y examin&oacute; las firmas moleculares de murci&eacute;lago humano aislado de m&uacute;ltiples dep&oacute;sitos adiposos. Para nuestra sorpresa, casi todos el murci&eacute;lago humano expresa abundantemente beige genes celulares selectivos, pero la expresi&oacute;n de genes de grasa parda selectivos cl&aacute;sicos eran casi imperceptibles. Curiosamente, expresi&oacute;n de los genes de grasa parda selectivos conocidos como PRDM16 se correlacion&oacute; fuertemente con eso de los genes celulares selectivos beige nuevamente identificados, pero no con el de genes selectivos de grasa marr&oacute;n cl&aacute;sicos. Adem&aacute;s, an&aacute;lisis histol&oacute;gicos demostraron que un nuevo marcador celular beige, CITED1, fue expresado selectivamente en lo positivo de la UCP1 beige c&eacute;lulas as&iacute; como en BAT humana. Brown adipose tissue (BAT) dissipates chemical energy and generates heat to protect animals from cold and obesity. Rodents possess two types of UCP-1 positive brown adipocytes arising from distinct developmental lineages: "classical" brown adipocytes develop during the prenatal stage whereas "beige" or "brite" cells that reside in white adipose tissue (WAT) develop during the postnatal stage in response to chronic cold or PPARÎ³ agonists. Beige cells' inducible characteristics make them a promising therapeutic target for obesity treatment, however, the relevance of this cell type in humans remains unknown. In the present study, we determined the gene signatures that were unique to classical brown adipocytes and to beige cells induced by a specific PPARÎ³ agonist rosiglitazone in mice. Subsequently we applied the transcriptional data to humans and examined the molecular signatures of human BAT isolated from multiple adipose depots. To our surprise, nearly all the human BAT abundantly expressed beige cell-selective genes, but the expression of classical brown fat-selective genes were nearly undetectable. Interestingly, expression of known brown fat-selective genes such as PRDM16 was strongly correlated with that of the newly identified beige cell-selective genes, but not with that of classical brown fat-selective genes. Furthermore, histological analyses showed that a new beige cell marker, CITED1, was selectively expressed in the UCP1-positive beige cells as well as in human BAT. These data indicate that human BAT may be primary composed of beige/brite cells.

Par la suite, nous avons appliqu&eacute; les donn&eacute;es transcriptional aux humains et examin&eacute; les signatures mol&eacute;culaires de l&#39;homme chauve-souris isol&eacute;s de plusieurs d&eacute;p&ocirc;ts adipeux. &Agrave; notre grande surprise, presque tous l&#39;homme chauve-souris des g&egrave;nes cellulaires-s&eacute;lectifs beiges abondamment exprim&eacute;s, mais l&#39;expression des g&egrave;nes de graisse brune s&eacute;lectifs classiques &eacute;taient presque ind&eacute;tectable. Fait int&eacute;ressant, l&#39;expression de g&egrave;nes de graisse brune s&eacute;lectifs connus comme PRDM16 est fortement corr&eacute;l&eacute;e avec celui des g&egrave;nes cellulaires-s&eacute;lectifs beiges nouvellement identifi&eacute;s, mais pas avec celle des g&egrave;nes s&eacute;lectifs de graisse brune classiques. En outre, des analyses histologiques ont montr&eacute; qu&#39;un nouveau marqueur de cellule beige, CITED1, a &eacute;t&eacute; s&eacute;lectivement exprim&eacute;e en l&#39;UCP1 s&eacute;ropositifs beiges cellules ainsi que dans l&#39;homme chauve-souris.
