We developed a three-dimensional assay prepared from primary breast cancer tissue and quantified tumor response to tamoxifen therapy. Freshly harvested breast cancer biopsies obtained at the time of curative surgical resection were fragmented and embedded into collagen I cushions. Changes in proliferation, apoptosis and tumor volume in response to tamoxifen treatment were quantified using image analysis software and optical projection tomography. Individual and collective invasion of epithelial cells into the surrounding collagen I was observed over the course of the experiment using phase contrast light microscopy and histopathological methods. Addition of tamoxifen to preparations derived from ER+ tumors demonstrated a range of response as measured by proliferative and apoptotic markers. In keeping with published data, tamoxifen reduced the percentage of apoptotic cells expressing cleaved caspase-3 (p = 0.02, Poisson regression analysis). Tamoxifen also reduced residual epithelial volume in ER+ tumors (p = 0.001, Mann-Whitney test), but not in ER low/- tumors (p = 0.78). Changes in tumor volume, as measured by optical projection tomography, allowed stratification into responsive and non-responsive tumors. The model mirrors observations of breast cancer response and histopathological changes to tamoxifen in neo-adjuvant trials. This assay provides a method of screening a battery of therapeutics against individual cancers, informing subsequent design of neo-adjuvant trials.

Determining tamoxifen sensitivity using primary breast cancer tissue in collagen-based three-dimensional culture.

Nous avons d&#xE9;velopp&#xE9; un test en trois dimensions pr&#xE9;par&#xE9; &#xE0; partir de tissus du cancer du sein primaire et de quantifier la r&#xE9;ponse tumorale &#xE0; un traitement au tamoxif&#xE8;ne. Fra&#xEE;chement r&#xE9;colt&#xE9;es biopsies de cancer du sein obtenus au moment de la r&#xE9;section chirurgicale curative ont &#xE9;t&#xE9; fragment&#xE9;es et int&#xE9;gr&#xE9;es dans du collag&#xE8;ne I coussins. Hemos desarrollado un ensayo en tres dimensiones preparado a partir de tejido primario del c&#xE1;ncer de mama y se cuantifica la respuesta del tumor a la terapia con tamoxifeno. Reci&#xE9;n cosechados, las biopsias de c&#xE1;ncer de mama obtenidas en el momento de la resecci&#xF3;n quir&#xFA;rgica curativa estaban fragmentados y embebido en col&#xE1;geno I cojines. Los cambios en el volumen de la proliferaci&#xF3;n, la apoptosis y el tumor en la respuesta al tratamiento con tamoxifeno se cuantificaron utilizando el software de an&#xE1;lisis de imagen y la tomograf&#xED;a por proyecci&#xF3;n &#xF3;ptica. La invasi&#xF3;n individual y colectiva de las c&#xE9;lulas epiteliales en el col&#xE1;geno que rodea I se observ&#xF3; en el transcurso del experimento con contraste de fase microscopio de luz y los m&#xE9;todos histopatol&#xF3;gicos. La adici&#xF3;n de tamoxifeno a las preparaciones derivadas de tumores ER + demostr&#xF3; una amplia gama de respuesta seg&#xFA;n lo medido por marcadores de proliferaci&#xF3;n y apoptosis. De acuerdo con los datos publicados, el tamoxifeno redujo el porcentaje de c&#xE9;lulas apopt&#xF3;ticas expresan exfoliados caspasa-3 (p = 0,02, el an&#xE1;lisis de regresi&#xF3;n de Poisson). El tamoxifeno tambi&#xE9;n redujo el volumen residual del epitelio en tumores ER + (p = 0,001, U de Mann-Whitney), pero no en baja / ER - los tumores (p = 0,78). Los cambios en el volumen del tumor, medidos mediante tomograf&#xED;a &#xF3;ptica de proyecci&#xF3;n, permiti&#xF3; la estratificaci&#xF3;n en los tumores sensibles y no sensibles. El modelo refleja las observaciones de la respuesta del c&#xE1;ncer de mama y los cambios histopatol&#xF3;gicos a tamoxifeno en el neo-adyuvante ensayos.
