Kaposi sarcoma (KS) is the most common AIDS-associated malignancy and is characterized by angiogenesis and the presence of spindle cells. Kaposi sarcoma-associated herpesvirus (KSHV) is consistently associated with all clinical forms of KS, and in vitro infection of dermal microvascular endothelial cells (DMVECs) with KSHV recapitulates many of the features of KS, including transformation, spindle cell proliferation, and angiogenesis. To study the molecular mechanisms of KSHV pathogenesis, we compared the protein expression profiles of KSHV-infected and uninfected DMVECs. This comparison revealed that heme oxygenase-1 (HO-1), the inducible enzyme responsible for the rate-limiting step in heme catabolism, was up-regulated in infected endothelial cells. Recent evidence suggests that the products of heme catabolism have important roles in endothelial cell biology, including apoptosis and angiogenesis. Here we show that HO-1 mRNA and protein are up-regulated in KSHV-infected cultures. Comparison of oral and cutaneous AIDS-KS tissues with normal tissues revealed that HO-1 mRNA and protein were also up-regulated in vivo. Increased HO-1 enzymatic activity in vitro enhanced proliferation of KSHV-infected DMVECs in the presence of free heme. Treatment with the HO-1 inhibitor chromium mesoporphyrin IX abolished heme-induced proliferation. These data suggest that HO-1 is a potential therapeutic target for KS that warrants further study.

Sarcome de Kaposi (SK) est la plus courante malignit&eacute; associ&eacute;es au sida et se caract&eacute;rise par l&#39;angiogen&egrave;se et la pr&eacute;sence de cellules de tige. Herp&egrave;svirus de sarcome-associ&eacute; de Kaposi (KSHV) est constamment associ&eacute; &agrave; toutes les formes cliniques de KS et infection in vitro de cellules endoth&eacute;liales microvasculaires (DMVECs) par voie cutan&eacute;e avec KSHV reprend bon nombre des caract&eacute;ristiques de KS, y compris la transformation, la prolif&eacute;ration de cellules fusiformes et l&#39;angiogen&egrave;se. Afin d&#39;&eacute;tudier les m&eacute;canismes mol&eacute;culaires de la pathog&eacute;nie KSHV, nous avons compar&eacute; les profils d&#39;expression de prot&eacute;ine de DMVECs KSHV-infect&eacute;s et non infect&eacute;s. Herpesvirus associato al sarcoma di Kaposi (KSHV) &egrave; costantemente associata a tutte le forme cliniche di KS, e infezione in vitro di dermale cellule endoteliali Microvascolari (DMVECs) con KSHV riassume molte delle caratteristiche di KS, compresa la trasformazione, la proliferazione cellulare del mandrino e l&#39;angiogenesi. Per studiare i meccanismi molecolari di patogenesi KSHV, abbiamo confrontato i profili di espressione della proteina di DMVECs KSHV-infetti e non infetti. Sarcoma de Kaposi (KS) es la neoplasia m&aacute;s frecuente asociado al SIDA y se caracteriza por la angiog&eacute;nesis y la presencia de c&eacute;lulas fusiformes. Herpesvirus asociado sarcoma de Kaposi (KSHV) se asocia sistem&aacute;ticamente a todas las formas cl&iacute;nicas de KS, y la infecci&oacute;n in vitro de c&eacute;lulas endoteliales microvasculares cut&aacute;neas (DMVECs) con KSHV recoge muchas de las caracter&iacute;sticas de KS, incluyendo la transformaci&oacute;n, la proliferaci&oacute;n de c&eacute;lulas fusiformes y angiog&eacute;nesis. Para estudiar los mecanismos moleculares de la patogenesia KSHV, comparamos los perfiles de expresi&oacute;n de la prote&iacute;na de DMVECs KSHV infectados y no infectados. Esta comparaci&oacute;n revel&oacute; que hemo oxigenasa-1 (HO-1), la enzima inducible responsable para el paso de limitaci&oacute;n de velocidad en el catabolismo del grupo heme, era regulado hasta en c&eacute;lulas endoteliales infectadas. La evidencia reciente sugiere que los productos del catabolismo del grupo heme tienen un papel importante en la biolog&iacute;a de la c&eacute;lula endotelial, incluyendo la apoptosis y la angiog&eacute;nesis. The inefficiency of lytic reactivation in various in vitro systems hampers the study of lytic genes in the context of whole virus. We report here increased expression of KSHV lytic genes and increased release of progeny virus when synchronized cultures of body cavity-based lymphoma-1 cells are treated with a phorbol ester during S phase of the cell cycle.

Kaposi's sarcoma-associated herpesvirus (KSHV) is involved in the development of lymphoproliferative diseases and Kaposi's sarcoma. The oncogenicity of this virus is reflected in vitro by its ability to transform B cells and endothelial cells. Infection of dermal microvascular endothelial cells (DMVEC) transforms the cells from a cobblestone-like monolayer to foci-forming spindle cells. This transformation is accompanied by dramatic changes in the cellular transcriptome. Known oncogenes, such as c-Kit, are among the KSHV-induced host genes. We previously showed that c-Kit is an essential cellular component of the KSHV-mediated transformation of DMVEC. Here, we test the hypothesis that the transformation process can be used to discover novel oncogenes. When expression of a panel of KSHV-induced cellular transcripts was inhibited with antisense oligomers, we observed inhibition of DMVEC proliferation and foci formation using antisense molecules to RDC1 and Neuritin. We further showed that transformation of KSHV-infected DMVEC was inhibited by small interfering RNA directed at RDC1 or Neuritin. Ectopic expression of Neuritin in NIH 3T3 cells resulted in changes in cell morphology and anchorage-independent growth, whereas RDC1 ectopic expression significantly increased cell proliferation. In addition, both RDC1- and Neuritin-expressing cells formed tumors in nude mice. RDC1 is an orphan G protein-coupled receptor, whereas Neuritin is a growth-promoting protein known to mediate neurite outgrowth. Neither gene has been previously implicated in tumorigenesis. Our data suggest that KSHV-mediated transformation involves exploitation of the hitherto unrealized oncogenic properties of RDC1 and Neuritin.

Novel cellular genes essential for transformation of endothelial cells by Kaposi's sarcoma-associated herpesvirus.

Herp&egrave;svirus associ&eacute; au sarcome de Kaposi (KSHV) est impliqu&eacute; dans le d&eacute;veloppement de maladies lymphoprolif&eacute;ratives et sarcome de Kaposi. L&#39;oncog&eacute;nicit&eacute; de ce virus se traduite in vitro par sa capacit&eacute; &agrave; transformer des cellules B et les cellules endoth&eacute;liales. L&#39;infection de cellules endoth&eacute;liales microvasculaires cutan&eacute;es (DMVEC) transforme les cellules d&#39;une monocouche pav&eacute;es ressemblant aux cellules de tige formant des foyers. Cette transformation s&#39;accompagne de changements dramatiques dans le transcriptome cellulaire. Oncog&egrave;nes connus, tels que le c-Kit, sont entre les g&egrave;nes de l&#39;h&ocirc;te KSHV-induite. Nous avons montr&eacute; pr&eacute;c&eacute;demment que c-Kit est une composante essentielle de cellulaire de la transformation de KSHV m&eacute;di&eacute;e par des DMVEC. L&#39;expression ectopique de Neuritin dans les cellules 3 t 3 NIH a entra&icirc;n&eacute; des changements dans la morphologie cellulaire et la croissance ind&eacute;pendante ancrage, consid&eacute;rant que l&#39;expression ectopique RDC1 a augment&eacute; consid&eacute;rablement la prolif&eacute;ration des cellules. En outre, les deux cellules exprimant RDC1 - et Neuritin forment des tumeurs chez la souris nude. RDC1 est un r&eacute;cepteur coupl&eacute; aux prot&eacute;ines G d&#39;orphelin, tandis que Neuritin est une prot&eacute;ine favorisant la croissance connue &agrave; la m&eacute;diation des neurites. Questa trasformazione &egrave; accompagnata da cambiamenti drammatici del transcriptome cellulare. Oncogeni noti, come il c-Kit, sono tra i geni host KSHV-indotta. Abbiamo precedentemente dimostrato che c-Kit &egrave; un componente cellulare essenziale della trasformazione KSHV-mediata del DMVEC. Qui, abbiamo testato l&#39;ipotesi che il processo di trasformazione pu&ograve; essere usato per scoprire nuovi oncogeni. Quando espressione di un pannello di trascrizioni cellulare indotta da KSHV &egrave; stato inibito con oligomeri antisenso, abbiamo osservato l&#39;inibizione della proliferazione di DMVEC e formazione di foci utilizzando molecole antisenso per RDC1 e Neuritin. Abbiamo ulteriormente dimostrato che trasformazione di DMVEC KSHV-infetti &egrave; stato inibito da piccolo RNA interferente diretto a RDC1 o Neuritin. Espressione ectopica di Neuritin in cellule 3T3 NIH ha provocato cambiamenti nella morfologia cellulare e la crescita ancoraggio-indipendente, mentre espressione ectopica RDC1 significativamente aumentata proliferazione cellulare. Inoltre, entrambi le cellule che esprimono RDC1 e Neuritin formano tumori nei topi nudi. RDC1 &egrave; un recettore orfano G protein-coupled, mentre Neuritin &egrave; una proteina growth-promoting nota per mediare escrescenza dei neuriti. N&eacute; gene ha precedentemente implicata nella tumorigenesi. Herpesvirus asociado sarcoma de Kaposi (KSHV) participa en el desarrollo de enfermedades linfoproliferativas y sarcoma de Kaposi. La oncogenicity de este virus se refleja in vitro por su capacidad para transformar las c&eacute;lulas B y c&eacute;lulas endoteliales. Infecci&oacute;n de c&eacute;lulas endoteliales microvasculares cut&aacute;neas (DMVEC) transforma las c&eacute;lulas de una monocapa de guijarro-como a la formaci&oacute;n de focos de c&eacute;lulas fusiformes. Esta transformaci&oacute;n se acompa&ntilde;a de cambios dram&aacute;ticos en el transcriptoma celular. Oncogenes conocidos, tales como c-Kit, son solo algunos de los genes del anfitri&oacute;n KSHV-inducida. Anteriormente mostramos que eso c-Kit es un componente esencial del celular de la transformaci&oacute;n mediada por KSHV de DMVEC. Aqu&iacute;, probamos la hip&oacute;tesis de que el proceso de transformaci&oacute;n puede ser usado para descubrir nuevos oncogenes. Cuando se inhibe la expresi&oacute;n de un panel de transcripciones celulares inducida por KSHV con olig&oacute;meros antisentidos, observamos la inhibici&oacute;n de la proliferaci&oacute;n de DMVEC y formaci&oacute;n de focos con mol&eacute;culas antisentidas para RDC1 y Neuritin. Expresi&oacute;n ect&oacute;pica de Neuritin en las c&eacute;lulas NIH 3T3 dio lugar a cambios en la morfolog&iacute;a celular y el crecimiento de anclaje independiente, mientras que la expresi&oacute;n ect&oacute;pica de RDC1 aument&oacute; significativamente la proliferaci&oacute;n celular. Adem&aacute;s, ambas c&eacute;lulas RDC1 - y Neuritin-expresi&oacute;n forman tumores en ratones desnudos. RDC1 es un receptor acoplado a prote&iacute;nas G del hu&eacute;rfano, mientras que Neuritin es una prote&iacute;na de crecimiento conocida para mediar consecuencia neurita. Ninguno de los dos genes se ha implicado previamente en la tumorig&eacute;nesis. Kaposi's sarcoma (KS) is a multifocal angioproliferative disorder affecting the skin, mucosa and viscera of individuals infected with human herpesvirus-8 (HHV-8; also Kaposi's sarcoma-associated herpesvirus [KSHV]). KS is the most common neoplasm in AIDS patients; the clinical outcome of AIDS-KS is significantly improved by highly active antiretroviral therapy (HAART). However, in Africa, where the severest manifestations of KS occur, there is limited access to these and other effective but expensive drugs. Here we present a review of current efforts to identify novel therapeutic targets for the treatment of KS using functional genomics, with recommendations regarding the development of economically feasible treatments for use in Africa.

Arcome de Kaposi (KS) est un trouble de l&#39;angioprolif&eacute;rative multifocale touchant la peau, les muqueuse et les visc&egrave;res des personnes infect&eacute;es par l&#39;herp&egrave;svirus humain-8 (HHV-8, &eacute;galement herp&egrave;svirus associ&eacute; au sarcome de Kaposi [KSHV]). KS est la tumeur plus courante chez les patients du sida ; le r&eacute;sultat clinique du sida-KS est nettement am&eacute;lior&eacute; par un traitement antir&eacute;troviral hautement actif (HAART). Cependant, en Afrique, o&ugrave; les manifestations les plus s&eacute;v&egrave;res de KS se produisent, il y a un acc&egrave;s limit&eacute; &agrave; ces et autre en vigueur mais les m&eacute;dicaments co&ucirc;teux. Sarcoma di Kaposi (KS) &egrave; una malattia multifocale angioproliferative che colpisce la pelle, mucosa e visceri delle persone infettate con herpesvirus-8 umano (HHV-8; anche associato al sarcoma di Kaposi herpesvirus [KSHV]). KS &egrave; la neoplasia pi&ugrave; comune nei pazienti con AIDS; l&#39;esito clinico del AIDS-KS &egrave; migliorato significativamente dalla terapia antiretrovirale altamente attiva (HAART). Sarcoma de Kaposi (KS) es un desorden de angioproliferative multifocal que afecta la piel, mucosa y v&iacute;sceras de los individuos infectados con herpesvirus-8 humano (HHV-8; tambi&eacute;n de sarcoma de Kaposi asociado herpesvirus [KSHV]). KS es la neoplasia m&aacute;s com&uacute;n en pacientes con SIDA; el resultado cl&iacute;nico de SIDA-KS se mejor&oacute; significativamente por la terapia antiretroviral altamente activa (HAART). Sin embargo, en &Aacute;frica, donde se producen las manifestaciones m&aacute;s severas de KS, hay acceso limitado a estos y otros efectivos pero costosos medicamentos. Aqu&iacute; presentamos una revisi&oacute;n de los actuales esfuerzos para identificar nuevas dianas terap&eacute;uticas para el tratamiento de KS utilizando la gen&oacute;mica funcional, con recomendaciones sobre el desarrollo de tratamientos econ&oacute;micamente factibles para uso en Africa.

Interfer&oacute;n (IFN)-&gamma; est&aacute; presente en lesiones de pacientes con la enfermedad de Lyme y se correlaciona positivamente con la severidad de las manifestaciones. Interferon (IFN)-Î³ is present in lesions of patients with Lyme disease and positively correlates with the severity of manifestations. To investigate the role of IFNÎ³ in the development of Lyme carditis, wild-type and IFNÎ³-deficient C57BL/6 mice were infected with the causative bacterium, Borrelia burgdorferi. Histological analysis revealed no change in the severity of carditis between wild-type and IFNÎ³-deficient mice at 14, 21, 25, and 28 days after infection. However, a distinct shift in the types of leukocytes within the hearts of IFNÎ³-deficient mice was observed at 25 days. In the absence of IFNÎ³, the number of neutrophils in the heart was increased, whereas the number of T lymphocytes was decreased. Bacterial loads within hearts were the same as in wild-type mice. Macrophages secrete chemokines that recruit immune cells, which could contribute to the accumulation of leukocytes in murine Lyme carditis. The ability of IFNÎ³ and B. burgdorferi to activate murine macrophages was examined, and the two stimuli synergistically induced chemoattractants for mononuclear cells (ie, CXCL9, CXCL10, CXCL11, CXCL16, and CCL12) and decreased those for neutrophils (ie, CXCL1, CXCL2, and CXCL3). IFNÎ³ and B. burgdorferi also synergistically enhanced secretion of CXCL9 and CXCL10 by murine cardiac endothelial cells. These results indicate that IFNÎ³ influences the composition of inflammatory infiltrates in Lyme carditis by promoting the accumulation of leukocytes associated with chronic inflammation and suppressing that of cells that typify acute inflammation.
