Neurogenese im postnatalen Gehirn hängt Wartung von drei biologischen Ereignisse: die Proliferation von Vorläuferzellen, die Migration von Neuroblasten, sowie die Differenzierung und Integration neuer Neuronen in bestehende neuronale Schaltkreise. Für die postnatale Neurogenese im Riechkolben, werden diese Ereignisse innerhalb von drei anatomisch verschiedene Domains getrennt: Verbreitung weitgehend erfolgt in der subependymale Zone (SEZ) der Seitenventrikel, Migration Neuroblasten durch den rostralen wandernden Strom (RMS) durchlaufen, und neue Neuronen zu differenzieren und Integration innerhalb der Riechkolben (OB). Die drei Domänen dienen als ideale Plattformen, um die zellulären, molekularen und physiologischen Mechanismen, die jedem der biologischen Ereignisse deutlich regulieren zu studieren. Dieses Papier beschreibt eine organotypischen Slice-Assay für die postnatale Hirngewebe optimiert, in denen die extrazelluläre Bedingungen genau imitieren die in vivo-Umgebung für die Migration von Neuroblasten. Wir zeigen, dass unser Assay für eine einheitliche, orientiert und schnelle Bewegung der Neuroblasten innerhalb der RMS bietet. Dieser Assay wird als sehr geeignet für die Untersuchung von Zell-autonome und nicht autonome Regulation neuronaler Migration durch die Nutzung Cross-Transplantation Ansätze von Mäusen auf unterschiedlichen genetischen Hintergründen.
