&#x827E;&#x6ECB;&#x75C5;&#x6BD2; Env &#x7ED1;&#x5B9A;&#x5230;&#x7531; 2 12 &#x6240;&#x63A2;&#x6D4B;&#x7684; Fc &#x6807;&#x8BB0; gp120 &#x5355;&#x514B;&#x9686;&#x6297;&#x4F53;&#x7684;&#x7EC6;&#x80DE;&#x53D7;&#x4F53;&#x7684;&#x6291;&#x5236;&#x4F5C;&#x7528;&#x3002;

During natural HIV infection, an array of host receptors are thought to influence virus attachment and the kinetics of infection. In this study, to probe the interactions of HIV envelope (Env) with various receptors, we assessed the inhibitory properties of various anti-Env monoclonal antibodies (mAbs) in binding assays. To assist in detecting Env in attachment assays, we generated Fc fusions of full-length wild-type gp120 and several variable loop-deleted gp120s. Through investigation of the inhibition of Env binding to cell lines expressing CD4, CCR5, DC-SIGN, syndecans or combinations thereof, we found that the broadly neutralizing mAb, 2G12, directed to a unique carbohydrate epitope of gp120, inhibited Env-CCR5 binding, partially inhibited Env-DC-SIGN binding, but had no effect on Env-syndecan association. Furthermore, 2G12 inhibited Env attachment to primary monocyte-derived dendritic cells, that expressed CD4 and CCR5 primary HIV receptors, as well as DC-SIGN, and suggested that the dual activities of 2G12 could be valuable in vivo for inhibiting initial virus dissemination and propagation.

Au cours de l&#39;infection naturelle par le VIH, un tableau des r&eacute;cepteurs de l&#39;h&ocirc;te sont cens&eacute;s influencer la fixation du virus et la cin&eacute;tique de l&#39;infection. Dans cette &eacute;tude, pour sonder les interactions de l&#39;enveloppe du VIH (Env) avec diff&eacute;rents r&eacute;cepteurs, nous avons &eacute;valu&eacute; les propri&eacute;t&eacute;s inhibitrices de divers anti-Env des anticorps monoclonaux (ACM) dans les tests de liaison. Pour aider &agrave; d&eacute;tecter les Env dans des essais de fixation, nous avons g&eacute;n&eacute;r&eacute; des fusions de Fc de pleine longueur sauvage gp120 et plusieurs gp120s variable de boucle-supprim&eacute;. Gr&acirc;ce &agrave; l&#39;enqu&ecirc;te de l&#39;inhibition de la liaison d&#39;Env aux lign&eacute;es de cellules exprimant CD4, CCR5, DC-SIGN, syndecans ou combinaisons de ces &eacute;l&eacute;ments, nous avons constat&eacute; que le mAb largement neutralisant, 12 2, r&eacute;alis&eacute; &agrave; un &eacute;pitope unique glucides de gp120, inhib&eacute;e la liaison Env-CCR5, partiellement inhib&eacute;e la liaison Env-DC-SIGN, mais n&#39;avait aucun effet sur l&#39;Env-syndecan association. W&auml;hrend nat&uuml;rliche HIV-Infektion ein Array von Host-Rezeptoren werden gedacht, um Virus-Anlage und die Kinetik der Infektion zu beeinflussen. Au&szlig;erdem hemmte 2 12 Env Anhaftung an prim&auml;ren Monozyte-abgeleitete dendritische Zellen, die CD4 und CCR5 prim&auml;re HIV-Rezeptoren als auch DC-SIGN ausgedr&uuml;ckt, und schlug vor, dualen Aktivit&auml;ten von 2-12 f&uuml;r Hemmung des urspr&uuml;nglichen Virus-Verbreitung und Vermehrung in Vivo wertvoll sein k&ouml;nnte.

Durante l&#39;infezione naturale da HIV, un array di recettori host sono pensato per influenzare l&#39;attacco di virus e la cinetica dell&#39;infezione. In questo studio, per sondare le interazioni della busta HIV (Env) con vari recettori, abbiamo valutato le propriet&agrave; inibitorie di vari anticorpi monoclonali anti-Env (mAbs) in associazione a dosaggi. Per aiutare nella rilevazione di Env in dosaggi di attaccamento, abbiamo generato Fc fusioni dell&#39;album wild-type gp120 e parecchie variabili loop-cancellato gp120s. Attraverso l&#39;indagine dell&#39;inibizione di Env associazione di linee cellulari che esprimono CD4, CCR5, DC-SIGN, syndecans o loro combinazioni, abbiamo trovato che il mAb ampiamente neutralizzante, 2 12, diretto a un epitopo unico carboidrato di gp120, inibito associazione Env-CCR5, parzialmente inibita associazione Env-DC-SIGN, ma ha avuto alcun effetto su Env-syndecan associazione. Inoltre, 12 2 inibito Env attaccamento alla primarie monocyte-derived cellule dendritiche, che esprime CD4 e CCR5 recettori HIV primari, cos&igrave; come DC-SIGN e ha suggerito che la doppia attivit&agrave; di 2 12 potrebbe essere preziosa in vivo per inibire la propagazione e la diffusione iniziale del virus.

Durante la infecci&oacute;n por VIH natural, matriz de receptores de host se piensan para influir en la fijaci&oacute;n del virus y la cin&eacute;tica de la infecci&oacute;n. En este estudio, para sondear las interacciones de la envoltura del VIH (Env) con diferentes receptores, se evaluaron las propiedades inhibitorias de varios anti-Env anticuerpos monoclonales (mAbs) en ensayos de Uni&oacute;n. Para ayudar en la detecci&oacute;n de Env en ensayos de accesorio, generamos Fc fusiones de larga duraci&oacute;n salvaje-tipo gp120 y varios gp120s variable de bucle-suprimido. A trav&eacute;s de la investigaci&oacute;n de la inhibici&oacute;n de la Uni&oacute;n de Env a l&iacute;neas celulares que expresan CD4, CCR5, DC-SIGN, sindecans o combinaciones de las mismas, encontramos que el mAb ampliamente neutralizante, 2 12, dirigido a un ep&iacute;tope &uacute;nico carbohidrato de gp120, inhibe Env-CCR5 vinculante, parcialmente inhibida vinculante Env-DC-SIGN, pero no tuvo efecto sobre la Asociaci&oacute;n Env-syndecan. Natural regulatory T cells (Tregs) are present in high frequencies among tumor-infiltrating lymphocytes and in draining lymph nodes, supposedly facilitating tumor development. To investigate their role in controlling local immune responses, we analyzed intratumoral T cell accumulation and function in the presence or absence of Tregs. Tumors that grew in normal BALB/c mice injected with the 4T1 tumor cell line were highly infiltrated by Tregs, CD4 and CD8 cells, all having unique characteristics. Most infiltrating Tregs expressed low levels of CD25Rs and Foxp3. They did not proliferate even in the presence of IL-2 but maintained a strong suppressor activity. CD4 T cells were profoundly anergic and CD8 T cell proliferation and cytotoxicity were severely impaired. Depletion of Tregs modified the characteristics of tumor infiltrates. Tumors were initially invaded by activated CD4(+)CD25(-) T cells, which produced IL-2 and IFN-gamma. This was followed by the recruitment of highly cytotoxic CD8(+) T cells at tumor sites leading to tumor rejection. The beneficial effect of Treg depletion in tumor regression was abrogated when CD4 helper cells were also depleted. These findings indicate that the massive infiltration of tumors by Tregs prevents the development of a successful helper response. The Tregs in our model prevent Th cell activation and subsequent development of efficient CD8 T cell activity required for the control of tumor growth.

Regulatory T cells prevent CD8 T cell maturation by inhibiting CD4 Th cells at tumor sites.

Les lymphocytes T r&eacute;gulateurs naturels (Tregs) sont pr&eacute;sents dans les hautes fr&eacute;quences chez les lymphocytes infiltrant &agrave; la tumeur et &agrave; drainer des ganglions lymphatiques, soi-disant facilitant le d&eacute;veloppement de tumeurs. Afin d&#39;&eacute;tudier leur r&ocirc;le dans le contr&ocirc;le des r&eacute;ponses immunitaires locales, nous avons analys&eacute; intratumorale accumulation de lymphocytes T et la fonction en pr&eacute;sence ou en absence de Tregs. Les tumeurs qui poussaient dans des souris BALB/c normales inject&eacute;s avec la lign&eacute;e de cellules tumorales 4 t 1 ont &eacute;t&eacute; fortement infiltr&eacute;s par des cellules Tregs, CD4 et CD8, tous ayant des caract&eacute;ristiques uniques. La plupart des Tregs infiltrantes exprim&eacute; low levels of CD25Rs and Foxp3. Ils ne pas prolif&eacute;rer m&ecirc;me en pr&eacute;sence de IL-2, mais maintient une activit&eacute; forte suppresseur. Les tumeurs &eacute;taient initialement envahis par les lymphocytes activ&eacute;s CD4(+)CD25(-) T, qui a produit des il-2 et l&#39;IFN-gamma. Ceci a &eacute;t&eacute; suivi par le recrutement de cellules hautement cytotoxiques CD8 T tumoraux menant au rejet de la tumeur. L&#39;effet b&eacute;n&eacute;fique de l&#39;appauvrissement de Treg dans une r&eacute;gression tumorale a &eacute;t&eacute; abrog&eacute;e lors de cellules auxiliaires CD4 sont &eacute;galement diminu&eacute;es. Ces r&eacute;sultats indiquent que l&#39;infiltration massive des tumeurs par Tregs emp&ecirc;che le d&eacute;veloppement d&#39;une intervention r&eacute;ussie d&#39;assistance. Les Tregs dans notre mod&egrave;le emp&ecirc;che l&#39;activation des lymphocytes Th et le d&eacute;veloppement subs&eacute;quent de l&#39;activit&eacute; des cellules T CD8 efficace n&eacute;cessaire pour le contr&ocirc;le de la croissance tumorale.

Nat&uuml;rliche regulatorische T-Zellen (Tregs) sind vorhanden in hohen Frequenzen zwischen den Tumor-infiltrieren Lymphozyten und Entw&auml;sserung Lymphknoten, Tumor-Entwicklung angeblich zu f&ouml;rdern. Um ihre Rolle bei der Kontrolle der lokalen Immunantworten zu untersuchen, analysiert wir intratumorale T-Zell-Akkumulation und Funktion im Vorhandensein oder Nichtvorhandensein von Tregs. Tumoren, die in normalen BALB/c M&auml;usen injiziert mit der 4T1 Tumor-Zelllinie wuchs stark infiltriert wurden Tregs, CD4 und CD8-Zellen, alle, die einzigartige Eigenschaften. Die meisten infiltrierendes Tregs ausgedr&uuml;ckt niedrige CD25Rs und Foxp3. Sie nicht vermehren sich auch in Gegenwart von Il aber eine starke Tumorsuppressor-Aktivit&auml;t beibehalten. CD4 T-Zellen waren zutiefst Anergic und CD8-T-Zellproliferation und Zytotoxizit&auml;t stark eingeschr&auml;nkt. Ersch&ouml;pfung der Tregs ver&auml;ndert die Eigenschaften der Tumor infiltriert. Tumoren wurden zun&auml;chst aktivierte CD4(+)CD25(-) T-Zellen, drangen die Il und IFN-Gamma produziert. Danach folgte die Einstellung von hoch zytotoxischen CD8(+) T-Zellen bei Tumor zu Tumor Ablehnung f&uuml;hrt. Die wohltuende Wirkung der Treg Ersch&ouml;pfung bei der Tumor-Regression wurde aufgehoben, wenn CD4-Helferzellen auch ersch&ouml;pft waren. Diese Befunde zeigen, dass das massive Eindringen von Tumoren von Tregs verhindert, die Entwicklung einer erfolgreichen Helfer-Antwort dass. Tregs in unserem Modell Th-ZELLAKTIVIERUNG und nachfolgende Entwicklung von effizienten CD8-T-Zellaktivit&auml;t ben&ouml;tigt, zur Steuerung von Tumorwachstum verhindern.

Regulatorische T-Zellen verhindern CD8-T-Zelle-Reifung durch Hemmung CD4 Th Zellen Tumor Standorte.

Naturale cellule T regolatorie (Tregs) sono presenti in alte frequenze tra linfociti tumore infiltrante e nel drenaggio dei linfonodi, presumibilmente facilitando lo sviluppo tumorale. Per indagare il loro ruolo nel controllo della risposta immunitaria locale, abbiamo analizzato intratumoral accumulo di cellule T e la funzione in presenza o assenza di Tregs. I tumori che crebbero in topi BALB/c normali iniettati con la linea di cellule tumorali 4T1 erano altamente infiltrati da cellule Tregs CD4 e CD8, tutti aventi caratteristiche uniche. La maggior parte dei Tregs infiltrante espresso bassi livelli di CD25Rs e di Foxp3. Non ha fatto proliferare anche in presenza di IL-2, ma mantenuto un&#39;attivit&agrave; forte soppressore. Le cellule T CD4 erano profondamente sommiers e citotossicit&agrave; e proliferazione delle cellule T CD8 erano gravemente compromesse. Deplezione di Tregs modificato le caratteristiche del tumore infiltrati. I tumori sono stati inizialmente invasa da cellule attivate T CD4(+)CD25(-), che ha prodotto il-2 e IFN-gamma. &#xADDC;&#xC81C; T &#xC138;&#xD3EC; &#xC885;&#xC591; &#xC0AC;&#xC774;&#xD2B8;&#xC5D0;&#xC11C; CD4 Th &#xC138;&#xD3EC;&#xB97C; &#xC5B5;&#xC81C; &#xD558; &#xC5EC; CD8 T &#xC138;&#xD3EC; &#xC131;&#xC219; &#xBC29;&#xC9C0;.

Las c&eacute;lulas T reguladoras naturales (Tregs) est&aacute;n presentes en altas frecuencias entre linfocitos infiltrantes de tumor y en el drenaje de los ganglios linf&aacute;ticos, supuestamente facilitar el desarrollo del tumor. Para investigar su papel en el control de la respuesta inmune local, hemos analizado intratumoral acumulaci&oacute;n de c&eacute;lulas T y la funci&oacute;n en la presencia o ausencia de Tregs. Tumores que crecieron en ratones BALB/c normales inyectados con la l&iacute;nea de c&eacute;lulas de tumor 4T1 eran altamente infiltrados por c&eacute;lulas Tregs, CD4 y CD8, teniendo caracter&iacute;sticas &uacute;nicas. Mayor&iacute;a Tregs infiltraci&oacute;n expres&oacute; bajos niveles de CD25Rs y Foxp3. No proliferar incluso en presencia de IL-2, pero mantuvo una actividad fuerte supresor. Las c&eacute;lulas T CD4 eran profundamente an&eacute;rgico y citotoxicidad y proliferaci&oacute;n de c&eacute;lulas T CD8 eran seriamente deterioradas. Agotamiento de Tregs modifica las caracter&iacute;sticas del tumor infiltra. Tumores inicialmente fueron invadidos por las c&eacute;lulas de T de CD4(+)CD25(-) activadas, que producen IL-2 y IFN-gamma. Esto fue seguido por el reclutamiento de c&eacute;lulas altamente citot&oacute;xicos de T CD8 en sitios de tumor conduce al rechazo del tumor. El efecto beneficioso de la depleci&oacute;n de Treg en la regresi&oacute;n del tumor fue abrogado cuando linfocitos CD4 tambi&eacute;n se agota. Estos resultados indican que la infiltraci&oacute;n masiva de tumores por Tregs impide el desarrollo de una respuesta acertada ayudante. Las Tregs en nuestro modelo de prevenir la activaci&oacute;n de las c&eacute;lulas Th y posterior desarrollo de la actividad de c&eacute;lulas T CD8 eficiente necesaria para el control del crecimiento tumoral.

Las c&eacute;lulas T reguladoras evitar la maduraci&oacute;n de c&eacute;lulas T CD8 mediante la inhibici&oacute;n de las c&eacute;lulas CD4 Th en sitios de tumor.
