The extracellular space (ECS) is the network of interconnected channels exterior to all brain cells and contains both interstitial fluid and extracellular matrix (Figure 1a and Figure 1b). The distribution of many substances required for brain cell function, including nutrients, hormones, and neurotransmitters, occurs by diffusion through the ECS. Changes in the physical parameters of this space, including volume, geometry, and extracellular matrix, can drastically affect diffusion through the ECS and the local ion concentrations bathing brain cells, which have a profound impact on brain cell function1,2.
Real-time iontophoresis (RTI) is used to determine two structural characteristics of a brain region: volume fraction and tortuosity3,4,5. Volume fraction (&#945;) is the proportion of tissue volume occupied by the ECS (VECS) relative to the total tissue volume (Vtissue) in a representative elementary volume;

Tortuosity (&#955;) is the relative hindrance that a substance encounters when diffusing through a brain region as compared to a medium with no obstructions;

where D* (cm2 s-1) is the effective diffusion coefficient of the substance in brain and D (cm2s-1) is the free diffusion coefficient of the substance in a free medium, such as dilute agarose gel.
Today, the most commonly used probe substance for the RTI method is the small cation tetramethylammonium (TMA). TMA has a molecular weight of 74 g/mol, completely dissociates in solution, and has one positive charge. RTI studies with this ion have demonstrated that &#945; <img alt="Equation" src="/files/ftp_upload/55755/55755eq3.jpg" /> 0.2 and &#955; <img alt="Equation" src="/files/ftp_upload/55755/55755eq3.jpg" /> 1.61,2. This means that the ECS is roughly 20% of the total brain volume and that the diffusion of a small, inert molecule occurs roughly 2.5 times slower in the ECS than in a medium with no obstructions3. However, both &#945; and &#955; vary with brain age, region, and state and in pathological conditions1. Alterations of these parameters have been linked to brain development, aging, sleep, epilepsy, and many other fundamental processes and diseases of the brain1,6. While other techniques measure &#945; and &#955;, RTI can measure both in localized regions of living tissue in real time. For this reason, RTI has become an indispensable tool for investigating changes in &#945; and &#955; during acute and reversible challenges.
The theory supporting RTI was originally validated by Nicholson and Phillips, and the technique has been used extensively since that time4,7. Experiments employing RTI begin with the release of a pulse of TMA from a source microelectrode by iontophoresis into a dilute agarose gel. Once ejected, the ions freely diffuse away from the point source, choosing from a potentially infinite number of random paths (Figure 1d). The changing concentration of the ion is measured over time using an ion-selective microelectrode (ISM) positioned roughly 100 &#181;m away (Figure 1c). The changes in TMA concentration are graphed and fitted to a curve that allows for the calculation of both D and the transport number of the iontophoresis microelectrode (parameters discussed in the Protocol). With these values, the procedure is repeated in a brain region of interest to obtain D* and to calculate both &#945; and &#955;. Control of the iontophoresis microelectrode, data collection, graphing and fitting of the TMA concentration curve, and calculation of the experimental parameters are all typically done by the programs Wanda and Walter, which have been specifically designed for this purpose (the software and their manuals are freely available from the authors upon request).
The Protocol section of this review describes the basic procedures needed to design and perform RTI in rodent brain slices. The technique has also been used in non-rodent models, including human brain slices and in vivo brain preparations1,4,6,8,9. The Representative Results section provides both ideal and non-ideal results to highlight nuances in data interpretation. Finally, the Discussion section briefly covers troubleshooting techniques, limitations of RTI, alternative techniques used to study the ECS, and future applications of RTI.

This review describes the basic concepts and protocol to perform the real-time iontophoresis (RTI) method, the gold-standard to explore and quantify the extracellular space (ECS) of the living brain. The ECS surrounds all brain cells and contains both interstitial fluid and extracellular matrix. The transport of many substances required for brain activity, including neurotransmitters, hormones, and nutrients, occurs by diffusion through the ECS. Changes in the volume and geometry of this space occur during normal brain processes, like sleep, and pathological conditions, like ischemia. However, the structure and regulation of brain ECS, particularly in diseased states, remains largely unexplored. The RTI method measures two physical parameters of living brain: volume fraction and tortuosity. Volume fraction is the proportion of tissue volume occupied by ECS. Tortuosity is a measure of the relative hindrance a substance encounters when diffusing through a brain region as compared to a medium with no obstructions. In RTI, an inert molecule is pulsed from a source microelectrode into the brain ECS. As molecules diffuse away from this source, the changing concentration of the ion is measured over time using an ion-selective microelectrode positioned roughly 100 &#181;m away. From the resulting diffusion curve, both volume fraction and tortuosity can be calculated. This technique has been used in brain slices from multiple species (including humans) and in vivo to study acute and chronic changes to ECS. Unlike other methods, RTI can be used to examine both reversible and irreversible changes to the brain ECS in real time.

The utility of the RTI technique is demonstrated in an experiment designed to measure the changes in &#945; and during a hypoosmolar challenge (Figure 8 and Figure 9). It has previously been shown that reducing the osmolarity of the ECS by washing on hypotonic ACSF will produce a decrease in &#945; and an increase in &#955;13.

Real-time Iontophoresis with Tetramethylammonium to Quantify Volume Fraction and Tortuosity of Brain Extracellular Space

This protocol describes real-time iontophoresis, a method that measures physical parameters of the extracellular space (ECS) of living brains. The diffusion of an inert molecule released into the ECS is used to calculate the ECS volume fraction and tortuosity. It is ideal for studying acute reversible changes to brain ECS.

The overall goal of this procedure is to quantify the volume fraction and tortuosity of the extracellular space in brain tissue. This method answers key questions regarding the relationship between the extracellular space and the physiology and disease. The main advantage of this technique is that it allows the real-time characterization of two important parameters of the extracellular space.Generally, individuals new to this method will struggle because the iontophoretic microelectrode exhibits a tendency to change its properties during experiments and frequently requires troubleshooting during experiments. Visual demonstration of this method is critical as the precise manipulation of an ion-selective microelectrode, an iontophoretic electrode over multiple steps is required to obtain a successful recording. To begin this procedure, run ACSF containing tetramethylammonium, or TMA, through the submersion chamber at two milliliters per minute.Set the temperature controller to a desired temperature and bubble the ACSF with 95%oxygen, 5%carbon dioxide for the duration of the experiment. Next, mount the ground electrode on a holder and submerge its tip into the ACSF running through the submersion chamber. Then, fill the porous cup with 0.3%agarose containing TMA prepared previously and place it in the submersion chamber.Now, secure an iontophoresis microelectrode to the pipette holder of one micromanipulator. Connect the iontophoresis microelectrode wire to the head stage of the iontophoresis unit. Next, secure a calibrated ISM to the second micromanipulator and connect the ISM to its head stages.Afterwards, turn on the computer containing the Wanda and Walter programs. In the Wanda GUI, click Calibrate. Then, in the Calibration box, fill in the voltages measured during the ISM calibration and click Fit Data.Following that, click Accept in the Calibrate box to automatically transfer the slope and interference generated to the main GUI. On the left side of the GUI, ensure that all the experimental parameters are set in the corresponding entries. Next, place a temperature probe in the agar cup.Record the measured temperature in the Temperature entry in the Measuring Electrode box of the GUI. Then, lower the microelectrodes at least 1, 000 micrometers deep into the agarose and center them in the cup. Visualize them under the microscope using a 10X objective.On the two-channel amplifier, manually move the ISM channel connector to the voltage subtraction output to set the subtraction on between the reference and ISM channels. Center the tips on each other in all three directional axes. Next, zero the relative positions of both microelectrodes on the micromanipulator control boxes and ensure that the microelectrodes are centered accurately and precisely.Then, move the ISM 120 micrometers away from the iontophoresis microelectrode in one axis. Subsequently, start a recording by clicking on Acquire in the GUI and allow the program to run for a full recording. In this procedure, open the Walter program on the computer.In the Zero Records From menu, click the Wanda Voltoro button to read the records generated by Wanda. In the next popup window, select one or more records to be read and click Open. In the One Write Excel menu, click Sheet 1.3 and follow prompts until the records are automatically graphed.To begin the fitting procedure, in the Two Options menu, click on the select rec button. Then, in the figure two popup window, use the mouse to move the cross hairs over the first record to be processed and press either mouse button to choose the record. After that, click on fit curve in the menu and use at least 20 iterations of fitting to obtain an accurate fit of the data.Next, select all to fit all data points and select continue. The program will fit the displayed curve. Then, select the option to write the result to the appropriate spreadsheet program by clicking Excel in the seven results menu.Name the Excel file. Note and record the data generated which will be used to determine the functionality of the iontophoresis microelectrode. If the iontophoresis microelectrode is deemed suitable for the experiment, record the average transport number from all trials in the TransportNum end field in the Wanda GUI.In this procedure, place a 400-micrometer-thick brain slice in the recording chamber ensuring that it is fully submerged in the flowing ACSF. Next, move both the iontophoresis microelectrode and the ISM above the field of interest on the brain slice. Submerge both electrodes in the flowing ACSF, but above the slice.Then, offset the voltage for both the reference and the ion-sensing channels to zero millivolts. Wait for the voltage in both channels to stabilize. On the chart recorder, mark the voltage measured on the ion-sensing channel of the ISM.Next, place the ISM and iontophoresis microelectrodes 200 micrometers deep in the slice and 120 micrometers away from each other. Calculate the voltage difference between the TMA signals measured in the flowing ACSF and in the brain and input this value into the Baseline Volt Millivolt field in the Measuring Electrode box of the Wanda GUI. On the left side of the GUI, ensure that all the experimental parameters are correctly entered.Start the recording by clicking Acquire and allow it to take a full recording. After moving both microelectrodes out of the slice, use the chart recorder to determine any change between the voltage measured now and its measurement from before. To analyze the recordings from the brain, repeat the procedures for agarose data analysis using the Walter program.Write the data onto the spreadsheet program by clicking Excel in the Walter menu. Then, record the volume fraction of the brain ECS, tortuosity of the brain ECS and the non-specific clearance. To check the transport number, take new recordings in agarose and repeat the procedures in the Walter program to obtain the transport number.Inspect the spreadsheet and check if the transport number has changed by more than 10%from the transport number obtained prior to the brain measurements. If so, the data obtained with the iontophoretic microelectrode are not reliable. The transport number measured in this step must closely match that obtained prior to brain measurements, otherwise the volume fraction cannot be considered accurate.Use newly obtained ISM calibration data as the input in the Wanda Calibrate box and check that the slope value differs by less than 10%from the previous calibration. Shown here is the representative data from a single trial obtained in agar demonstrating the concentration curve of TMA. And this is a fitted curve obtained from data processing in Walter.The overlap of the data in the fitted curve demonstrates that the curve fitting done by Walter accurately models diffusion in this trial. Here is the table of agar measurements before experimentation in the brain. Here is a summary of measurements taken in a mouse brain slice under the control and experimental conditions.Alpha decreased during a hypoosmolar challenge while tortuosity increased as expected. Alpha overshot the control values in the recovery conditions. And here is the table of agra measurements after experimentation in the brain.The average transport numbers before and after the experiment were within 10%of each other which indicate that the values recorded in the brain were reliable. Once mastered, this procedure can be performed in 14 hours over two days. On day one, ISMs are fabricated and calibrated.On day two, ISMs are re-calibrated and real-time iontophoresis is performed. While attempting this procedure, it's important to first be precise in the spacing of your electrodes. Secondly, it's important to only accept an experiment if the transport number of the iontophoretic microelectrode remains stable throughout the course of the experiment.Following this procedure, which characterizes the diffusion of small cations in the extracellular space, you can go onto to perform other techniques like the integrative optical imaging method that allows you to answer the question of how large micromolecules move in the extracellular space. After watching this video, you should have a good understanding of how to perform the real-time iontophoresis method to quantify brain extracellular space volume and tortuosity in living brain in health and disease. Fast Micro-iontophoresis of Glutamate and GABA: A Useful Tool to Investigate Synaptic Integration

One of the fundamental interests in neuroscience is to understand the integration of excitatory and inhibitory inputs along the very complex structure of the dendritic tree, which eventually leads to neuronal output of action potentials at the axon. The influence of diverse spatial and temporal parameters of specific synaptic input on neuronal output is currently under investigation, e.g. the distance-dependent attenuation of dendritic inputs, the location-dependent interaction of spatially segregated inputs, the influence of GABAergig inhibition on excitatory integration, linear and non-linear integration modes, and many more.
With fast micro-iontophoresis of glutamate and GABA it is possible to precisely investigate the spatial and temporal integration of glutamatergic excitation and GABAergic inhibition. Critical technical requirements are either a triggered fluorescent lamp, light-emitting diode (LED), or a two-photon scanning microscope to visualize dendritic branches without introducing significant photo-damage of the tissue. Furthermore, it is very important to have a micro-iontophoresis amplifier that allows for fast capacitance compensation of high resistance pipettes. Another crucial point is that no transmitter is involuntarily released by the pipette during the experiment.
Once established, this technique will give reliable and reproducible signals with a high neurotransmitter and location specificity. Compared to glutamate and GABA uncaging, fast iontophoresis allows using both transmitters at the same time but at very distant locations without limitation to the field of view. There are also advantages compared to focal electrical stimulation of axons: with micro-iontophoresis the location of the input site is definitely known and it is sure that only the neurotransmitter of interest is released. However it has to be considered that with micro-iontophoresis only the postsynapse is activated and presynaptic aspects of neurotransmitter release are not resolved. In this article we demonstrate how to set up micro-iontophoresis in brain slice experiments.

Real-time Imaging of Axonal Transport of Quantum Dot-labeled BDNF in Primary Neurons

BDNF plays an important role in several facets of neuronal survival, differentiation, and function. Structural and functional deficits in axons are increasingly viewed as an early feature of neurodegenerative diseases, including Alzheimer&#8217;s disease (AD) and Huntington&#8217;s disease (HD). As yet unclear is the mechanism(s) by which axonal injury is induced. We reported the development of a novel technique to produce biologically active, monobiotinylated BDNF (mBtBDNF) that can be used to trace axonal transport of BDNF. Quantum dot-labeled BDNF (QD-BDNF) was produced by conjugating quantum dot 655 to mBtBDNF. A microfluidic device was used to isolate axons from neuron cell bodies. Addition of QD-BDNF to the axonal compartment allowed live imaging of BDNF transport in axons. We demonstrated that QD-BDNF moved essentially exclusively retrogradely, with very few pauses, at a moving velocity of around 1.06 &#956;m/sec. This system can be used to investigate mechanisms of disrupted axonal function in AD or HD, as well as other degenerative disorders.

Axonal transport of BDNF, a neurotrophic factor, is critical for the survival and function of several neuronal populations. Some degenerative disorders are marked by disruption of axonal structure and function. An Ex Vivo Laser-induced Spinal Cord Injury Model to Assess Mechanisms of Axonal Degeneration in Real-time

Injured CNS axons fail to regenerate and often retract away from the injury site. Axons spared from the initial injury may later undergo secondary axonal degeneration. Lack of growth cone formation, regeneration, and loss of additional myelinated axonal projections within the spinal cord greatly limits neurological recovery following injury. To assess how central myelinated axons of the spinal cord respond to injury, we developed an ex vivo living spinal cord model utilizing transgenic mice that express yellow fluorescent protein in axons and a focal and highly reproducible laser-induced spinal cord injury to document the fate of axons and myelin (lipophilic fluorescent dye Nile Red) over time using two-photon excitation time-lapse microscopy. Dynamic processes such as acute axonal injury, axonal retraction, and myelin degeneration are best studied in real-time. However, the non-focal nature of contusion-based injuries and movement artifacts encountered during in vivo spinal cord imaging make differentiating primary and secondary axonal injury responses using high resolution microscopy challenging. The ex vivo spinal cord model described here mimics several aspects of clinically relevant contusion/compression-induced axonal pathologies including axonal swelling, spheroid formation, axonal transection, and peri-axonal swelling providing a useful model to study these dynamic processes in real-time. Major advantages of this model are excellent spatiotemporal resolution that allows differentiation between the primary insult that directly injures axons and secondary injury mechanisms; controlled infusion of reagents directly to the perfusate bathing the cord; precise alterations of the environmental milieu (e.g., calcium, sodium ions, known contributors to axonal injury, but near impossible to manipulate in vivo); and murine models also offer an advantage as they provide an opportunity to visualize and manipulate genetically identified cell populations and subcellular structures. Here, we describe how to isolate and image the living spinal cord from mice to capture dynamics of acute axonal injury.

We present a protocol utilizing two-photon excitation time-lapse microscopy to simultaneously visualize the dynamics of axon and myelin injuries in real time. Real-time Electrophysiology: Using Closed-loop Protocols to Probe Neuronal Dynamics and Beyond

Experimental neuroscience is witnessing an increased interest in the development and application of novel and often complex, closed-loop protocols, where the stimulus applied depends in real-time on the response of the system. Recent applications range from the implementation of virtual reality systems for studying motor responses both in mice1 and in zebrafish2, to control of seizures following cortical stroke using optogenetics3. A key advantage of closed-loop techniques resides in the capability of probing higher dimensional properties that are not directly accessible or that depend on multiple variables, such as neuronal excitability4 and reliability, while at the same time maximizing the experimental throughput. In this contribution and in the context of cellular electrophysiology, we describe how to apply a variety of closed-loop protocols to the study of the response properties of pyramidal cortical neurons, recorded intracellularly with the patch clamp technique in acute brain slices from the somatosensory cortex of juvenile rats. As no commercially available or open source software provides all the features required for efficiently performing the experiments described here, a new software toolbox called LCG5 was developed, whose modular structure maximizes reuse of computer code and facilitates the implementation of novel experimental paradigms. Stimulation waveforms are specified using a compact meta-description and full experimental protocols are described in text-based configuration files. Additionally, LCG has a command-line interface that is suited for repetition of trials and automation of experimental protocols.

A Protocol for the Administration of Real-Time fMRI Neurofeedback Training

Neurologic disorders are characterized by abnormal cellular-, molecular-, and circuit-level functions in the brain. New methods to induce and control neuroplastic processes and correct abnormal function, or even shift functions from damaged tissue to physiologically healthy brain regions, hold the potential to dramatically improve overall health. Of the current neuroplastic interventions in development, neurofeedback training (NFT) from functional Magnetic Resonance Imaging (fMRI) has the advantages of being completely non-invasive, non-pharmacologic, and spatially localized to target brain regions, as well as having no known side effects. Furthermore, NFT techniques, initially developed using fMRI, can often be translated to exercises that can be performed outside of the scanner without the aid of medical professionals or sophisticated medical equipment. In fMRI NFT, the fMRI signal is measured from specific regions of the brain, processed, and presented to the participant in real-time. Through training, self-directed mental processing techniques, that regulate this signal and its underlying neurophysiologic correlates, are developed. FMRI NFT has been used to train volitional control over a wide range of brain regions with implications for several different cognitive, behavioral, and motor systems. Additionally, fMRI NFT has shown promise in a broad range of applications such as the treatment of neurologic disorders and the augmentation of baseline human performance. In this article, we present an fMRI NFT protocol developed at our institution for modulation of both healthy and abnormal brain function, as well as examples of using the method to target both cognitive and auditory regions of the brain.

Assay to Measure Nucleocytoplasmic Transport in Real Time within Motor Neuron-like NSC-34 Cells

Nucleocytoplasmic transport refers to the import and export of large molecules from the cell nucleus. Recently, a number of studies have shown a connection between amyotrophic lateral sclerosis (ALS) and impairments in the nucleocytoplasmic pathway. ALS is a neurodegenerative disease affecting the motor neurons and resulting in paralysis and ultimately in death, within 2-5 years on average. Most cases of ALS are sporadic, lacking any apparent genetic linkage, but 10% are inherited in a dominant manner. Recently, hexanucleotide repeat expansions (HREs) in the chromosome 9 open reading frame 72 (C9orf72) gene were identified as a genetic cause of ALS and frontotemporal dementia (FTD). Importantly, different groups have recently proposed that these mutants affect nucleocytoplasmic transport. These studies have mostly shown the final outcome and manifestations caused by HREs on nucleocytoplasmic transport, but they do not demonstrate nuclear transport dysfunction in real time. As a result, only severe nucleocytoplasmic transport deficiency can be determined, mostly due to high overexpression or exogenous protein insertion. 
This protocol describes a new and very sensitive assay to evaluate and quantify nucleocytoplasmic transport dysfunction in real time. The rate of import of a NLS-NES-GFP protein (shuttle-GFP) can be quantified in real time using fluorescent microscopy. This is performed by using an exportin inhibitor, thus allowing the shuttle GFP only to enter the nucleus. To validate the assay, the C9orf72 HRE translated dipeptide repeats, poly(GR) and poly(PR), which have been previously shown to disrupt nucleocytoplasmic transport, were used. Using the described assay, a 50% decrease in the nuclear import rate was observed compared to the control. Using this system, minute changes in nucleocytoplasmic transport can be examined and the ability of different factors to rescue (even partially) a nucleocytoplasmic transport defect can be determined.

Live-cell Measurement of Odorant Receptor Activation Using a Real-time cAMP Assay

The enormous sizes of the mammalian odorant receptor (OR) families present difficulties to find their cognate ligands among numerous volatile chemicals. To efficiently and accurately deorphanize ORs, we combine the use of a heterologous cell line to express mammalian ORs and a genetically modified biosensor plasmid to measure cAMP production downstream of OR activation in real time. This assay can be used to screen odorants against ORs and vice versa. Positive odorant-receptor interactions from the screens can be subsequently confirmed by testing against various odor concentrations, generating concentration-response curves. Here we used this method to perform a high-throughput screening of an odorous compound against a human OR library expressed in Hana3A cells and confirmed that the positively-responding receptor is the cognate receptor for the compound of interest. We found this high-throughput detection method to be efficient and reliable in assessing OR activation and our data provide an example of its potential use in OR functional studies.

Recording Synaptic Plasticity in Acute Hippocampal Slices Maintained in a Small-volume Recycling-, Perfusion-, and Submersion-type Chamber System

Even though experiments on brain slices have been in use since 1951, problems remain that reduce the probability of achieving a stable and successful analysis of synaptic transmission modulation when performing field potential or intracellular recordings. This manuscript describes methodological aspects that might be helpful in improving experimental conditions for the maintenance of acute brain slices and for recording field excitatory postsynaptic potentials in a commercially available submersion chamber with an outflow-carbogenation unit. The outflow-carbogenation helps to stabilize the oxygen level in experiments that rely on the recycling of a small buffer reservoir to enhance the cost-efficiency of drug experiments. In addition, the manuscript presents representative experiments that examine the effects of different carbogenation modes and stimulation paradigms on the activity-dependent synaptic plasticity of synaptic transmission.

Exploring Deep Space - Uncovering the Anatomy of Periventricular Structures to Reveal the Lateral Ventricles of the Human Brain

Anatomy students are typically provided with two-dimensional (2D) sections and images when studying cerebral ventricular anatomy and students find this challenging. Because the ventricles are negative spaces located deep within the brain, the only way to understand their anatomy is by appreciating their boundaries formed by related structures. Looking at a 2D representation of these spaces, in any of the cardinal planes, will not enable visualisation of all of the structures that form the boundaries of the ventricles. Thus, using 2D sections alone requires students to compute their own mental image of the 3D ventricular spaces. The aim of this study was to develop a reproducible method for dissecting the human brain to create an educational resource to enhance student understanding of the intricate relationships between the ventricles and periventricular structures. To achieve this, we created a video resource that features a step-by-step guide using a fiber dissection method to reveal the lateral and third ventricles together with the closely related limbic system and basal ganglia structures. One of the advantages of this method is that it enables delineation of the white matter tracts that are difficult to distinguish using other dissection techniques. This video is accompanied by a written protocol that provides a systematic description of the process to aid in the reproduction of the brain dissection. This package offers a valuable anatomy teaching resource for educators and students alike. By following these instructions educators can create teaching resources and students can be guided to produce their own brain dissection as a hands-on practical activity. We recommend that this video guide be incorporated into neuroanatomy teaching to enhance student understanding of the morphology and clinical relevance of the ventricles.

In Vivo Microdialysis Method to Collect Large Extracellular Proteins from Brain Interstitial Fluid with High-molecular Weight Cut-off Probes

In vivo microdialysis is a powerful technique to collect ISF from awake, freely-behaving animals based on a dialysis principle. While microdialysis is an established method that measures relatively small molecules including amino acids or neurotransmitters, it has been recently used to also assess dynamics of larger molecules in ISF using probes with high molecular weight cut off membranes. Upon using such probes, microdialysis has to be run in a push-pull mode to avoid pressure accumulated inside of the probes. This article provides step-by-step protocols including stereotaxic surgery and how to set up microdialysis lines to collect proteins from ISF. During microdialysis, drugs can be administered either systemically or by direct infusion into ISF. Reverse microdialysis is a technique to directly infuse compounds into ISF. Inclusion of drugs in the microdialysis perfusion buffer allows them to diffuse into ISF through the probes while simultaneously collecting ISF. By measuring tau protein as an example, the author shows how its levels are altered upon stimulating neuronal activity by reverse microdialysis of picrotoxin. Advantages and limitations of microdialysis are described along with the extended application by combining other in vivo methods.
