G -タンパク質共役受容体（GPCR）は7回膜貫通型タンパク質ファミリーに属し、細胞内応答する細胞外シグナルの伝達を媒介する。 GPCRの制御などの走化性、細胞内カルシウムの放出、ヘテロ三量体G -タンパク質1から2を介してリガンド依存的に遺伝子の調節など、多様な生物学的機能。リガンドの結合は、そのようなサイクリックアデノシン一リン酸（cAMP）、イノシトール三リン酸（IP3）とジアシルグリセロール（DG）などのセカンドメッセンジャーのレベルを調節するヘテロ三量体G -タンパク質の活性化につながる立体構造変化のシリーズを誘導する。受容体のリガンド結合の活性化との併用はまた脱感作、隔離および/または内在化を介したシグナル受容体を減衰させる一連のイベントを開始します。 GPCRの脱感作のプロセスは、G -タンパク質の受容体キナーゼ（GRKs）とβ-アレスチン3のその後の結合による受容体のリン酸化を介して行われます。 β-アレスチンは、細胞質タンパク質であり、受容体の内在化4-6容易にすることで、そこに（ほとんどの場合）リン酸化受容体に結合し、GPCRの活性化によって細胞膜に転位する。 とLTB 4受容体2（BLT2、低親和性受容体、ロイコトリエンB 4（LTB 4）GPCRの、LTB 4受容体1（高親和性受容体BLT1）を介してアラキドン酸経路と媒介する、その行動から派生した炎症誘発性脂質分子である）7-9。 LTB 4 - BLT1経路は、喘息、関節炎、アテローム性動脈硬化症10-17日 、など、いくつかの炎症性疾患において重要であることが示されている。現在の紙は、LTB 4誘発性白血球遊走およびβ-アレスチンと、顕微鏡イメージング技術の18から19を用いて生細胞における受容体の転座を持つBLT1の相互作用を監視するために開発した方法論を説明します。 C57BL / 6マウスから樹状細胞を骨髄由来は、前述の20〜21として単離し、培養した。これらの細胞は、LTB 4による細胞遊走を示すために、生細胞イメージング法で試験した。人間のBLT1は、C -末端に赤色蛍光タンパク質（BLT1 - RFP）を付けられ、β- arrestin1は、緑色蛍光タンパク質（β- ARR - GFP）で標識し、ラット好塩基球Leukomia（RBL - 2H3）細胞株に両方のプラスミドをトランスフェクトした18-19これらのタンパク質とローカリゼーション間の相互作用のカイネティクスは、ライブセルビデオ顕微鏡を用いてモニターした。現在の論文の方法論は、生細胞におけるGタンパク質共役型受容体の機能的応答を調べるために顕微鏡技術の使用を説明しています。現在の紙はまた、受容体の動態と細胞質蛋白質の相互作用を決定するために蛍光強度を定量化するMetamorphソフトウェアの使用を説明します。
