<ol><li> <strong>Opzetten van filtratie apparatuur</strong> <strong></strong><ol><li> Voor het instellen van het filtreerapparaat, gebruik een pincet in een klasse GF / D-filter te plaatsen in het voorfilter wooneenheid. Losjes vast de behuizing met de hand. Als een vuistregel, niet te strak of het apparaat zal lekken zodra de pompen begint. </li><li> Vervang de mandfles dop met de geautoclaveerde dop en de bijgevoegde slang door zorgvuldig het inbrengen van de interne slang in de mandfles, zonder aan te raken. <p class="jove_content"> Het is noodzakelijk om te vegen van banken en pompen met een vochtige handdoek na gebruik aan een slepende sporen van zout water die anders zouden aantasten apparatuur te verwijderen. Hoewel de procedure is eenvoudig, kan de benodigde tijd uitgebreid worden afhankelijk van de concentratie van biomassa of afval in het monster wordt gefilterd. </p>

<p class="jove_content">De workflow begint met het verzamelen van de kust mariene wateren voor de downstream microbiële gemeenschap, nutriënten-en trace gas analyses. Voor deze methode werden monsters verzameld vanaf het dek van de HMS John Strickland die actief zijn in Saanich Inlet. Deze video documenten groot volume (≥ 20 L) filtratie van microbiële biomassa, variërend van 0.22μm en 2.7μm in diameter, uit het water kolom. Twee 20L monsters kunnen gelijktijdig worden gefilterd met een enkele pomp unit uitgerust met vier draaibare koppen. Filtratie is gedaan in het veld op lange reizen, of direct bij terugkeer voor dagtochten. Het is belangrijk om op te nemen van de hoeveelheid water die door elke sterivex filter unit. Om te voorkomen dat biofilmvorming tussen de monsterneming reizen, moeten alle filtratie-apparatuur worden gespoeld met verdund HCl en gedeïoniseerd water en geautoclaveerd onmiddellijk na gebruik. Deze procedure duurt ongeveer 5 uur, plus een extra uur voor het opruimen.</p>
