<p class="jove_content"> Het is noodzakelijk om te vegen van banken en pompen met een vochtige handdoek na gebruik aan een slepende sporen van zout water die anders zouden aantasten apparatuur te verwijderen. Hoewel de procedure is eenvoudig, kan de benodigde tijd uitgebreid worden afhankelijk van de concentratie van biomassa of afval in het monster wordt gefilterd. </p>

<p class="jove_content">De workflow begint met het verzamelen van de kust mariene wateren voor de downstream microbiële gemeenschap, nutriënten-en trace gas analyses. Voor de demonstratie van vandaag en monsters werden verzameld vanaf het dek van de HMS John Strickland die actief zijn in Saanich Inlet. Deze video documenten klein volume (~ 1 L) filtratie van microbiële biomassa uit de waterkolom. Het protocol is een uitbreiding van het grote volume bemonsteringsprotocol eerder beschreven, met een groot verschil: hier is er geen pre-filtratie stap, zodat alle grootteklassen van biomassa worden verzameld beneden de 0,22 urn filter cut-off. Monsters op deze manier zijn ideaal voor het nucleïnezuur analyse. De set-up, filtratie en clean-up stappen ieder voor zich ongeveer 20-30 minuten. Als u twee peristaltische pompen tegelijk, maximaal 8 monsters kunnen worden gefilterd op hetzelfde moment. Om te voorkomen dat biofilmvorming tussen de monsterneming reizen, moeten alle filtratie-apparatuur worden gespoeld met verdund HCl en gedeïoniseerd water en geautoclaveerd onmiddellijk na gebruik.</p>
