es cell-derived neuroepithelial cell cultures

ES-cellen hebben de potentie om te differentiëren in de cellen van alle kiembladen, waardoor ze een aantrekkelijk instrument voor de ontwikkeling van nieuwe therapieën. In het algemeen is de differentiatie van ES-cellen volgt het concept om eerst te genereren onvolgroeide stamcellen, die vervolgens kunnen worden uitgedragen en gedifferentieerd tot volgroeide cellulaire fenotypes. Dit geldt ook voor de ES-cel afgeleide neurogenese, waarin de ontwikkeling van zenuwcellen volgt twee belangrijke stappen: Ten eerste, de afleiding en de uitbreiding van onvolwassen neuro precursoren en seconden, hun differentiatie tot volwassen neurale cellen. Een veel voorkomende methode om neurale voorlopercellen te produceren uit ES-cellen is gebaseerd op embryoid lichaam (EB) formatie, waarin de differentiatie van cellen uit alle kiembladen inclusief neuroectoderm onthult. Een alternatief en meer efficiënte methode om neuro-epitheliale cel ontwikkeling te induceren maakt gebruik van stromale cel afkomstige inducerende activiteit (SDIA), die kan worden bereikt door co-kweken van embryonale stamcellen met de schedel beenmerg-afgeleide stromale cellen (1). Beide, EB vorming en SDIA, onthullen de ontwikkeling van de rozet-achtige structuren, die worden verondersteld om neurale buis lijken op-en / of neurale crest-achtige voorouders. De neurale voorlopers kunnen worden geïsoleerd, uitgebreid en verder gedifferentieerd in specifieke neuronen en glia cellen met behulp van bepaalde kweekomstandigheden. We beschrijven hier de generatie en het isolement van deze rozetten in co-cultuur van experimenten met de stromale cellijn MS5 (2-5).
