measuring replicative life span in the budding yeast

El envejecimiento es un proceso degenerativo caracterizado por un deterioro progresivo de los componentes celulares y orgánulos que resulta en la mortalidad. La levadura en ciernes<em&gt; Saccharomyces cerevisiae</em&gt; Se ha utilizado ampliamente para estudiar la biología del envejecimiento, y hay varios factores determinantes de la longevidad de la levadura se ha demostrado que se conserva en eucariotas multicelulares, incluyendo gusanos, moscas y ratones<sup&gt; 1</sup&gt;. Aquí se presenta un protocolo generalizado para medir la duración de la vida replicativa de las células de levadura en ciernes madre. El objetivo del ensayo de replicación vida es determinar cuántas veces los brotes de células madre. Las células madre y la hija se pueden distinguir fácilmente por un investigador experimentado con un microscopio óptico estándar (un aumento total de 160X), como el Zeiss Axioscope 40 u otro modelo similar. La separación física de las células hijas a partir de células madre se realiza mediante un micromanipulador manuales equipado con una aguja de fibra óptica. Típico de las cepas de levadura de laboratorio produce 20 a 30 células hijas por madre y un experimento de vida requiere de 2-3 semanas.
