<p class="jove_content">In de klassieke eiwitkleuring protocollen met behulp van Coomassie Brilliant Blue (CBB), oplossingen met een hoog gehalte aan giftige en brandbare organische oplosmiddelen (methanol, ethanol of 2-propanol) en azijnzuur worden gebruikt voor fixatie, kleuren en ontkleuren van eiwitten in een gel na SDS -PAGE. Het versnellen van de procedure, het verwarmen van de kleuroplossing in de magnetron voor een korte tijd wordt vaak gebruikt. Dit resulteert meestal in verdamping van giftige of gevaarlijke methanol, ethanol of 2-propanol en een sterke geur van azijnzuur in het lab die moeten worden vermeden vanwege de veiligheid overwegingen. In een protocol oorspronkelijk gepubliceerd in twee octrooiaanvragen door EM Wondrak (US2001046709 (A1), US6319720 (B1)), is een alternatieve samenstelling van de kleuroplossing beschreven waarin geen organische oplosmiddelen of zuur wordt gebruikt. Het CBB is opgelost in dubbelgedestilleerd water (60-80 mg van het CBB G-250 per liter) en 35 mM HCl wordt toegevoegd als de enige andere verbinding in de kleuroplossing. Het CBB staning van de gel gebeurt na SDS-PAGE en grondig wassen van de gel in dubbelgedestilleerd water. Door het verwarmen van de gel tijdens het wassen en vlekken stappen, kan het proces sneller worden afgewerkt en er geen giftige of gevaarlijke compunds worden verdampen. De kleuring van eiwitten gebeurt al binnen 1 minuut na het verwarmen van de gel in kleuroplossing en is volledig ontwikkeld na 15-30 minuten met een licht blauwe achtergrond die volledig ontkleurd bij langdurige wassen van de gekleurde gel in dubbelgedestilleerd water, zonder dat de lood-eiwit bands.</p>
