Dit protocol is relatief eenvoudig, maar elke stap de praktijk nodig heeft om te zorgen dat de embryo&#39;s niet beschadigd zijn. Dit protocol beschrijft het gebruik van de Drosophila melanogaster syncytieel embryo&#39;s voor het bestuderen van mitose 1. Drosophila is nuttig genetica met een gesequenced genoom, en kan eenvoudig worden onderhouden en gemanipuleerd. Veel mitotische mutanten bestaan, en transgene vliegen uiten van functionele fluorescent (bijv. GFP) - tagged mitotische eiwitten zijn en worden gegenereerd. Gerichte genexpressie is mogelijk met de GAL4/UAS systeem 2. De Drosophila vroege embryo voert meerdere mitoses zeer snel (celcyclus duur, ≈ 10 min). Het is zeer geschikt voor de beeldvorming mitose, want tijdens de cycli 10-13, kernen verdelen snel en synchroon zonder in te grijpen cytokinese op het oppervlak van het embryo in een monolaag net onder de cortex. Deze snel delende kernen waarschijnlijk gebruik maken van dezelfde machines als mitotische andere cellen, maar ze zijn geoptimaliseerd voor snelheid, het ijkpunt wordt algemeen aangenomen dat niet te streng, waardoor de studie van de mitotische eiwitten wier afwezigheid zou de celcyclus veroorzaken in cellen met een sterke checkpoint . Embryo&#39;s uitdrukken GFP gelabelde eiwitten of gemicroinjecteerd met fluorescent gelabelde eiwitten kunnen eenvoudig worden gescand om te leven dynamiek (fig. 1) te volgen. Daarnaast kunnen embryo&#39;s worden gemicroinjecteerd met functie-blokkerende antilichamen of remmers van specifieke eiwitten om het effect van het verlies of verstoring van hun functie 3 studie. Deze reagentia kan diffunderen door het embryo, het bereiken van een groot aantal spindels met een helling van de concentratie van de remmer, die op zijn beurt resulteert in een gradiënt van gebreken die vergelijkbaar zijn met een allelische reeks van mutanten. Productie In het ideale geval als het doel eiwit fluorescent gelabeld is, kan de gradiënt van remming direct gevisualiseerd 4. Aangenomen wordt dat de sterkste fenotype is vergelijkbaar met de nul-fenotype, ook al is het moeilijk om formeel uitgesloten dat de antilichamen kunnen dominante effecten in zeldzame gevallen, zo strenge controle en voorzichtige interpretatie hebben moeten worden toegepast. Verder weg van de plaats van injectie, eiwit-functie is slechts gedeeltelijk verloren gaat waardoor andere functies van het doelwit eiwit aan zichtbaar worden.
