Detta protokoll används idag i vårt laboratorium och har förfinats under årens lopp av många människor, inklusive Dr David Sharp, Mijung Kwon, Patrizia Sommi, Dhanya Cheerambathur. Detta protokoll är relativt enkelt, men kräver att varje steg i praktiken att se till att embryona inte är skadade. Detta protokoll beskriver användningen av Drosophila melanogaster syncytial embryo för att studera mitos 1. Drosophila har bra genetik med ett ordnat genomet, och det kan vara lätt att underhålla och manipuleras. Många mitotiska mutanter existerar och transgena flugor uttrycka funktionella fluorescerande (t.ex. GFP) - taggade mitotiska proteiner har och håller på att skapas. Riktade genuttryck är möjlig med GAL4/UAS system 2. Den Drosophila tidiga embryot utför flera mitoses mycket snabbt (cellcykeln varaktighet, ≈ 10 min). Det är väl lämpad för bildhantering mitos, eftersom det under cykler 10-13, kärnor delar snabbt och synkront utan att ingripa cytokines på ytan av embryot i en enda monolager precis under cortex. Dessa snabbt delande kärnor använder antagligen samma mitotiska maskiner som andra celler, men de är optimerade för hastighet, riktpunkten är allmänt tros inte vara stränga, vilket gör att studien av mitotiska proteiner vars frånvaro skulle orsaka gripandet cellcykeln i celler med en stark checkpoint . Embryon som uttrycker GFP märkta proteiner eller idag injiceras med fluorescerande proteiner kan lätt avbildas följa leva dynamik (Fig. 1). Dessutom kan embryon idag injiceras med funktion-blockerande antikroppar eller hämmare av specifika proteiner för att studera effekten av förlust eller störning av deras funktion 3. Dessa reagenser kan sprida i hela embryot, nå många spindlar för att producera en lutning på koncentration av hämmare, som i sin tur resulterar i en gradient av defekter kan jämföras med en allela serie av mutanter. Helst om målprotein fluorescerande är märkt, kan lutning på inhibition direkt visualiseras 4. Det förutsätts att den starkaste fenotypen är jämförbart med noll fenotypen, men det är svårt att formellt utesluta att antikroppar kan ha dominerande effekter i sällsynta fall, så en noggrann kontroll och försiktig tolkning måste tillämpas. Längre bort från injektionsstället är proteiners funktion endast delvis förlorat låta andra funktioner målprotein för att bli uppenbara.
