La tomographie optique de projection est une technique d&#39;imagerie en trois dimensions qui a été récemment présenté comme un outil d&#39;imagerie principalement dans la biologie du développement et les études d&#39;expression génique. La technique rend échantillon biologique optiquement transparents en commençant par les déshydrater et en plaçant ensuite dans un mélange d&#39;alcool benzylique et de benzoate de benzyle dans un rapport 2:1 (BABB ou Murray solution S Clair). La technique rend les échantillons biologiques optiquement transparents en commençant par les déshydratant dans des solutions d&#39;éthanol à les placer ensuite dans un mélange d&#39;alcool benzylique et de benzoate de benzyle dans un rapport 2:1 (ou une solution claire BABB Murray s) à effacer. Après le processus de compensation de la contribution de diffusion dans l&#39;échantillon peut être considérablement réduit et a fait presque négligeable alors que la contribution d&#39;absorption ne peut pas être complètement éliminés. En essayant de reconstruire la distribution de fluorescence dans l&#39;échantillon de l&#39;enquête, cette contribution affecte les reconstructions et conduit inévitablement à des artefacts d&#39;image et des erreurs de quantification .. Alors que l&#39;absorption pourrait être encore réduit avec une permanence de plusieurs semaines ou mois dans les médias de compensation, ce qui conduira à une perte progressive de la fluorescence et à un temps de traitement des échantillons anormalement long. Cela est vrai lorsque la reconstruction de deux agents de contraste exogènes (agents de contraste moléculaires) ainsi que le contraste endogène (par exemple des reconstitutions génétiquement exprimé protéines fluorescentes).
