Hoewel de menselijke embryonale stamcellen (hESC) is aangetoond dat een stabiele diploïde karyotype 1 aanwezig is, hebben vele studies blijkt dat, afhankelijk van de kweekomstandigheden ze gevoelig zijn voor chromosomale afwijkingen zoals het toevoegen van geheel of delen van de chromosomen te verwerven. Inderdaad, tijdens langdurige cultuur, karyotypic veranderingen waargenomen als enzymatische of chemische dissociatie worden gebruikt 2,3,4, terwijl de handleiding dissectie van kolonies voor de passage behoudt een stabiele karyotype 5. Trouwens, veranderingen in de omgeving, zoals het verwijderen van feeder-cellen lijken ook de genetische integriteit van hESC 3,6 compromis. Zodra chromosomale veranderingen van invloed kunnen zijn cellulaire fysiologie, van cruciaal belang gezien hESC is het karakteriseren van de genetische integriteit van hESC in vitro als een essentieel instrument in de embryogenese studies en drugs testen. Verder is het voor de toekomstige therapeutische doeleinden chromosomale veranderingen zijn een echte zorg omdat het vaak is gekoppeld aan kanker. Hier laten we een eenvoudige en handige methode om een hoge kwaliteit chromosoom spreads te verkrijgen voor verdere analyse van de chromosomen die door G-banding, FISH, lucht of de CGH technieken 7,8. Wij raden u aan de chromosomale-status routinematig met tussenpozen van vijf passages in om het uiterlijk van translocaties en aneuploïdieën te controleren Priscila Britto en Rafaela Sartore droegen evenzeer voor het papier.
