We hopen dat deze video demonstratie heeft onze methoden weergegeven voor live-embryo dissectie en kleuring in voldoende detail dat ze nu gemakkelijk kan worden uitgevoerd door een persoon die enige ervaring heeft in Drosophila genetica en embryologie. Natuurlijk zullen veel praktijk nog steeds nodig zijn, vooral voor de dissecties zelf. Drosophila embryo&#39;s tussen de etappes 14 en 17 van de embryonale ontwikkeling kan gemakkelijk worden ontleed om &quot;filet&quot; preparaten te genereren. In deze voorbereidingen, het centrale zenuwstelsel loopt in het midden, en wordt geflankeerd door het lichaam muren. Veel verschillende fenotypes zijn onderzocht met behulp van een dergelijke preparaten. In de meeste gevallen werden de filets gegenereerd door ontleding van antilichaam-gekleurde vaste hele-mount embryo&#39;s. Deze &quot;vaste dissecties&quot; hebben een aantal nadelen, echter. Ze zijn tijdrovend uit te voeren, en het is moeilijk om mutant (GFP-negatief) embryo&#39;s sorteren van voorraden waarin mutaties in de loop der GFP balancer chromosomen. Sinds 2002 is onze groep is het uitvoeren van een tekort en ectopische expressie schermen om liganden voor weesreceptoren te identificeren. Om dit te doen, we ontwikkelden gestroomlijnde protocollen voor de live-embryo dissectie en antilichaam kleuring van collecties met honderden gebalanceerde lijnen. We hebben geconcludeerd dat het aanzienlijk efficiënter om fenotypes in grote collecties van de voorraden te onderzoeken door live dissectie dan door vaste dissectie. Met behulp van het protocol hier beschreven, kan een enkel individu opgeleid tot het scherm tot 10 lijnen per dag voor fenotypes, onderzoek 4-7 mutant embryo&#39;s van elke regel onder een samengestelde microscoop. Dit maakt de identificatie van mutaties die een subtiele, lage penetrantie fenotypes, omdat tot 70 hemisegments per lijn worden gescoord bij een sterke vergroting met een 40X water-immersie lens.
