<p class="jove_content">Herkömmliche optische Mikroskope Bildzellen durch die Verwendung von Objektiven, die zusammen mit anderen Objektiven und optischen Komponenten. Während sehr effektiv, hat diese klassische Vorgehensweise bestimmte Einschränkungen für die Miniaturisierung der Imaging-Plattform kompatibel zu machen mit den fortgeschrittenen Stand der Technik in der Mikrofluidik. In diesem Bericht stellen wir experimentelle Details eines ohne Linsen auskommt, On-Chip-Imaging-Konzept bezeichnet LUCAS (<strong<em&gt; L</em</strong&gt; Ensless<strong<em&gt; U</em</strong&gt; LTRA-weites Feld-of-view<strong<em&gt; C</em</strong&gt; Ell Überwachung<strong<em&gt; A</em</strong&gt; Rray Plattform basierend auf<strong<em&gt; S</em</strong&gt; Hadow imaging), die benötigt keine Mikroskop-Objektive oder andere sperrige optische Komponenten zum Bild einer heterogenen Zell-Lösung über einen extrem weiten Bereich der Ansicht, dass span so groß wie kann ~ 18 cm<sup&gt; 2</sup&gt;. Darüber hinaus, im Gegensatz zu herkömmlichen Mikroskopen können LUCAS Bild einer heterogenen Zell-Lösung von Interesse über eine Tiefe-of-Bereich ~ 5 mm, ohne die Notwendigkeit für eine Neuausrichtung, die bis ~ 9 mL Probenvolumen entspricht. Das Imaging-Plattform zeichnet die Schatten (dh ohne Linsen auskommt, digitale Hologramme) jeder Zelle von Interesse in seinem Blickfeld, und automatisierte digitale Verarbeitung dieser Zelle Schatten kann der Typ, der Graf und die relativen Positionen von Zellen in der Lösung zu bestimmen. Weil es erfordert kein sperriges optischen Komponenten oder mechanischen Scanningtische bietet es eine deutlich miniaturisierte Plattform, die gleichzeitig reduziert die Kosten, die ganz wichtig ist vor allem Point of Care Diagnostik. Darüber hinaus ist die Bildgebung Durchsatz von dieser Plattform Größenordnungen besser als bei herkömmlichen optischen Mikroskopen, die als überaus wertvoll für die Hochdurchsatz-Zell-Biologie Experimente könnten.</p>
