O ser humano no trato gastrointestinal (GI) é um ambiente único em que células do epitélio intestinal e não-patogênicos (comensais) bactérias coexistem. Tem sido proposto que o microambiente que o patógeno encontra na camada de comensais é importante para determinar a extensão da colonização. Métodos de cultura atual para investigar a colonização de patógenos não são adequados para investigar esta hipótese, pois não permitem co-cultura de bactérias e células epiteliais de uma forma que imita o microambiente do trato GI. Descrevemos aqui um modelo de co-cultura microfluídicos que permite que a cultura independente de células eucarióticas e bactérias, e testar o efeito do microambiente comensais sobre a colonização do patógeno. O modelo de co-cultura é demonstrada através do desenvolvimento de um comensal<em&gt; Escherichia coli</em&gt; Biofilme entre as células HeLa, seguido pela introdução de entero<em&gt; E. coli</em&gt; (EHEC) na ilha comensais, em uma seqüência que imita a seqüência de eventos em infecção do trato GI.
