<p class="jove_content"> Angiogenes är en komplicerad process i flera steg, där som svar på angiogena stimuli, nya fartyg skapas från den befintliga kärlbädden. Dessa åtgärder inkluderar: nedbrytning av basalmembranet, spridning och migration (spirande) av endotelceller (EG) i den extracellulära matrisen, anpassning av EG i sladdar, lumen bildning, anastomos och bildandet av en ny basalmembranet. Många in vitro-tester har utvecklats för att studera denna process, men de flesta bara härma vissa skeden av angiogenes, och morfologiskt fartygen ofta inte liknar fartygen in vivo. Här visar vi ett optimerat in vitro-angiogenes test som använder mänskliga EG navelsträngen ven och fibroblaster. Denna modell rekapitulerar alla de viktigaste tidiga stadierna av angiogenes, och viktigast fartygen displayen patentet intercellulära lumen omgiven av polariserad EG. Fartyg kan lätt observeras av faskontrast och Time-lapse mikroskopi och som återvinns i ren form för tillämpningar i efterföljande led. </p>
