<td>10 g/bottle. 
1.	0.5 g of dry beads are hydrated and swollen in 50 mL PBS (pH=7.4) for at least 3 hours at RT.
Use a 50 mL tube and place it on the rocker.
2.	Let the beads settle down (~ 15 min). Discard the supernatant and wash the beads for a few minutes in fresh PBS (50 mL).
3.	Discard the PBS and replace with fresh PBS:
25 mL -> 20 mg/mL => 60000 beads/mL or
50 mL -> 10 mg/mL => 30000 beads/ mL
4.	Place the bead suspension in a siliconized glass bottle (Windshield Wiper or Sigmacote).
5.	Sterilize the beads by autoclaving for 15 min at 115&#xF0B0;C.
6.	Store it at 4&#xF0B0;C.

optimized fibrin gel bead assay for the study of angiogenesis

<p class="jove_content">L&#39;angiogenèse est un complexe multi-étapes, où, en réponse aux stimuli angiogéniques, les nouveaux navires sont créés à partir de la vascularisation existante. Ces étapes comprennent: la dégradation de la membrane basale, la prolifération et la migration (germination) des cellules endothéliales (CE) dans la matrice extracellulaire, l&#39;alignement de la CE en cordons, de branchement, la formation de lumière, anastomose, et la formation d&#39;une nouvelle membrane basale. Beaucoup d&#39;essais in vitro ont été développées pour étudier ce processus, mais la plupart ne reproduisent certaines étapes de l&#39;angiogenèse, et morphologiquement les vaisseaux dans les dosages ne sont souvent pas ressembler à des navires in vivo. Basé sur des travaux antérieurs et Nehls Drenckhahn, nous avons optimisé un dosage de l&#39;angiogenèse in vitro qui utilise la veine ombilicale humaine CE et les fibroblastes. Ce modèle reprend toutes les étapes clés au début de l&#39;angiogenèse et, surtout, les vaisseaux d&#39;affichage des brevets lumens intercellulaire entouré par polarisée CE. CE sont enduites sur microporteurs Cytodex et intégrés dans un gel de fibrine. Les fibroblastes sont en couches sur le dessus du gel, où ils assurent nécessaires facteurs solubles qui favorisent la germination CE de la surface des billes. Après plusieurs jours, de nombreux navires sont actuellement qui peut facilement être observée sous contraste de phase et de la microscopie time-lapse. Cette vidéo montre les étapes clés à mettre en place ces cultures.</p>
