<td>10 g/bottle. 
1.	0.5 g of dry beads are hydrated and swollen in 50 mL PBS (pH=7.4) for at least 3 hours at RT.
Use a 50 mL tube and place it on the rocker.
2.	Let the beads settle down (~ 15 min). Discard the supernatant and wash the beads for a few minutes in fresh PBS (50 mL).
3.	Discard the PBS and replace with fresh PBS:
25 mL -> 20 mg/mL => 60000 beads/mL or
50 mL -> 10 mg/mL => 30000 beads/ mL
4.	Place the bead suspension in a siliconized glass bottle (Windshield Wiper or Sigmacote).
5.	Sterilize the beads by autoclaving for 15 min at 115&#xF0B0;C.
6.	Store it at 4&#xF0B0;C.

optimized fibrin gel bead assay for the study of angiogenesis

<p class="jove_content">Angiogenes är en komplicerad process i flera steg, där, som svar på angiogena stimuli, nya fartyg skapas från den befintliga kärlbädden. Dessa åtgärder inkluderar: nedbrytning av basalmembranet, spridning och migration (spirande) av endotelceller (EG) i den extracellulära matrisen, anpassning av EG i sladdar, förgrening, lumen bildning, anastomos och bildandet av en ny basalmembranet. Många in vitro-tester har utvecklats för att studera denna process, men de flesta bara härma vissa skeden av angiogenes, och morfologiskt fartygen i analyserna ofta inte liknar fartygen in vivo. Baserat på tidigare arbeten av Nehls och Drenckhahn, har vi optimerat en in vitro-angiogenes test som använder mänskliga EG navelsträngen ven och fibroblaster. Denna modell rekapitulerar alla de viktigaste tidiga stadierna av angiogenes och, viktigare, fartyg visas patentet intercellulära lumen omgiven av polariserad EG. EG är belagda på cytodex microcarriers och inbäddad i en fibrin gel. Fibroblaster är skiktade ovanpå gelen där de ger nödvändiga lösliga faktorer som främjar EG groning från ytan av pärlorna. Efter flera dagar, många fartyg är närvarande som lätt kan observeras i faskontrast och time-lapse mikroskopi. Denna video visar de viktigaste stegen för att inrätta dessa kulturer.</p>
