<p class="jove_content">Omdat de video-data zijn complex en bestaan ​​uit veel afbeeldingen, mijnbouw informatie van videomateriaal is moeilijk om te doen zonder de hulp van de computer software. Video bioinformatica is een krachtige kwantitatieve benadering voor de extractie van ruimte-tijd gegevens van video-beelden met behulp van computer software voor dating mining en analyse uit te voeren. In dit artikel introduceren we een video bioinformatica methode voor het kwantificeren van de groei van menselijke embryonale stamcellen (hESC) door het analyseren van time-lapse video&#39;s verzameld in een Nikon BioStation CT-incubator uitgerust met een camera voor video-imaging. In onze experimenten werden hESC kolonies die werden vastgemaakt aan Matrigel gefilmd voor 48 uur in de BioStation CT. Voor het bepalen van het tempo van de groei van deze kolonies werden recepten ontwikkeld met behulp van CL-Quant software die gebruikers in staat stelt te extraheren verschillende soorten van gegevens van videobeelden. Om nauwkeurig te evalueren kolonie groei werden drie recepten gecreëerd. De eerste gesegmenteerd het beeld in de kolonie en de achtergrond, de tweede verbeterde het beeld om kolonies nauwkeurig te definiëren in de video-sequentie, en de derde gemeten van het aantal pixels in de kolonie in de tijd. De drie recepten werden uitgevoerd in de juiste volgorde op de video-data verzameld in een BioStation CT om de snelheid van de groei van individuele hESC kolonies meer dan 48 uur te analyseren. Te controleren of de waarheid van de CL-Quant recepten, werden dezelfde gegevens handmatig geanalyseerd met behulp van Adobe Photoshop-software. Wanneer de gegevens verkregen met behulp van de CL-Quant recepten en Photoshop werden vergeleken waren de resultaten nagenoeg identiek zijn, met vermelding van de CL-Quant recepten eerlijk waren. De hier beschreven methode kan worden toegepast op alle video-gegevens groeipercentages van hESC of andere cellen die groeien in kolonies te meten. Daarnaast kunnen andere video bioinformatica recepten worden ontwikkeld in de toekomst voor andere cel processen, zoals migratie, apoptose, en celadhesie.</p>
