Le termiti sono tra i pochi animali noti per avere la capacità di sussistere esclusivamente da legno consumando. Il tratto intestinale termite contiene una popolazione microbica densi e ricchi di specie che assiste nella degradazione della lignocellulosa prevalentemente in acetato, la sostanza nutriente chiave alimentando il metabolismo delle termiti (Odelson &amp; Breznak, 1983). All&#39;interno di queste popolazioni microbiche sono batteri, archeobatteri metanogeni e, in alcuni (&quot;inferiore&quot;) termiti, eucarioti protozoi. La composizione delle specie delle popolazioni microbiche in budella delle termiti così come i geni chiave coinvolti in vari processi biochimici è stata esplorata mediante tecniche molecolari (Kudo et al, 1998;. Schmit-Wagner et al, 2003;. Salmassi &amp; Leadbetter, 2003). Queste tecniche dipendono dalla estrazione e purificazione degli acidi nucleici di elevata qualità dall&#39;ambiente intestinale termite. La tecnica di estrazione descritta in questo video è una raccolta modificata di protocolli sviluppati per l&#39;estrazione e la purificazione degli acidi nucleici da campioni ambientali (Mor et al, 1994;. Berthelet et al, 1996;. Purdy et al, 1996;. Salmassi &amp; Leadbetter, 2003;.
