in situ quantification of pancreatic beta-cell mass in mice

Отслеживание изменений конкретных популяций клеток в норме и патологии является важной целью проведения биомедицинских исследований. Процесс мониторинга панкреатических бета-клеток островков и рост особенно сложным. Мы разработали метод захвата распределения бета-клеток в поджелудочной железе нетронутыми трансгенных мышей с флуоресцентными метками бета-клеток с макроса, написанного для ImageJ (rsb.info.nih.gov / ц /). После вскрытия поджелудочной железы и ткани очистки, всей поджелудочной железы захватывается в качестве виртуального ломтик, после чего GFP-меткой бета-клеток рассматриваются. Анализ включает в себя количественную оценку суммарной бета-клеток площадь, количество островок и распределение по размерам со ссылкой на конкретные параметры и места для каждого островка и для небольших кластеров бета-клеток. Все распределение островков могут быть построены в трех измерениях, а также информации от распространения на размер и форму каждого островка позволяет количественное и качественное сравнение изменений в общей бета-клеток области с первого взгляда.
