Neuronen aus dem Gehirn von embryonalen / postnatale Nagetiere geerntet werden in primären Zellkulturen gezüchtet werden, wo sie Neuriten und erweitern Form funktioneller synaptischer Verbindungen. Coating Deckgläser: Put autoklaviert Deckgläschen in einer 60 mm Petrischale. Pipettieren Sie 5 ul der Con A / Laminin-Mix auf Mitte jedes Deckglas. Legen Sie in 37 C Inkubator für 2 Stunden. Spülen Sie 3x Deckgläschen mit 100 ul autoklaviert Wasser jeden. Verwenden Sie einen Staubsauger an einer sterilen Pasteur-Pipette. Während der letzten Spülgang, nehmen Sie den Deckglas mit einer Pinzette und trocken beiden Seiten. Übertragen Sie die Deckglas zu einer 35-mm-Petrischale. Lagerung bei Raumtemperatur für bis zu einem Monat.

In diesem Video zeigen wir die Herstellung von primären neuronalen Kulturen aus den Gehirnen von Spätstadium Drosophila Puppen. Das Verfahren beginnt mit der Entfernung der Gehirne von Tieren, bei 70-78 Std. nach Puparium Bildung. Die isolierte Gehirne sind nach kurzer Inkubationszeit in Papain von mehreren Wäschen in serumfreien Wachstumsmedium gefolgt gezeigt. Der Prozess der mechanischen Dissoziation jedes Gehirn in einem 5-ul Tropfen Medien auf einem Deckglas dargestellt. Die Axone und Dendriten der post-mitotischen Neuronen off in der Nähe des Soma während Dissoziation sheered aber die Neuronen beginnen, um Prozesse innerhalb von wenigen Stunden plating regenerieren. Bilder zeigen, lebende Kulturen in 2 Tagen. Neuronen weiterhin Prozesse während der ersten Woche in Kultur zu erarbeiten. Speziellen neuronalen Populationen können in Kultur identifiziert werden mit GAL4 Linien gewebespezifische Expression von fluoreszierenden Markern wie GFP oder RFP-Laufwerk. Ganzzell-Aufnahmen haben gezeigt, den kultivierten Neuronen bilden funktionelle, spontan aktive cholinergen und GABAergen Synapsen. Ein kurzes Video Segment zeigt Calcium-Dynamik in der kultivierten Neuronen mit Fura-2 als Kalzium-Indikator-Farbstoff zu spontanen Kalzium Transienten und Nikotin hervorgerufenen Calcium-Reaktionen in eine Schüssel mit kultivierten Neuronen zu überwachen. Diese Puppen Gehirn Kulturen sind ein nützliches Modell, bei dem genetische und pharmakologische Werkzeuge zur inneren und äußeren Faktoren, die Bildung und Funktion von zentralen Synapsen beeinflussen identifiziert werden können.
