Per iniziare, prendere una fiaschetta di cellule di fibroblasti in coltura, rivestire una piastra di coltura cellulare di 10 centimetri adatta per la coltura hiPSC con quattro millilitri di soluzione di rivestimento a freddo per piastra e incubare la piastra per un'ora a 37 gradi Celsius. Successivamente, rimuovere il mezzo dai fibroblasti e lavarli con quattro millilitri di PBS preriscaldato per pallone. Aspirare il PBS e associare i fibroblasti con quattro millilitri di tripsina-EDTA per pallone incubando per cinque-sette minuti a 37 gradi Celsius.
Monitorare il distacco delle cellule utilizzando un microscopio a contrasto di fase e, se necessario, picchiettare contro il lato del pallone per staccare le cellule dalla superficie di plastica. Trasferire le cellule staccate in un tubo conico da 50 millilitri e lavare i palloni vuoti con sei millilitri di mezzo fibroblastico preriscaldato. Raccogliere e raggruppare le celle rimanenti nel tubo conico.
Quindi centrifugare le cellule a 200 g per cinque minuti a 20 gradi Celsius. Aspirare il surnatante e risospendere il pellet cellulare in tre millilitri di PBS fresco preriscaldato. Contare le cellule, trasferire 1,5 volte 10 al sesto fibroblasto in un nuovo tubo conico e riempire con PBS fino al segno di cinque millimetri.
Centrifugare il tubo a 200 g per cinque minuti a 20 gradi Celsius. Scartare il surnatante, risospendere il pellet cellulare in cinque millilitri di mezzo di elettroporazione e centrifugare nuovamente. Nel frattempo, aspirare la soluzione di rivestimento dalla piastra di coltura cellulare rivestita da 10 centimetri e aggiungere sette millilitri di terreno fibroblastico preriscaldato.
Al termine della centrifugazione, scartare il surnatante e risospendere il pellet cellulare in mezzo di elettroporazione con una concentrazione di 1,5 volte 10 alla sesta cella in 250 microlitri. Trasferire la sospensione cellulare da 250 microlitri in una cuvetta di elettroporazione con una distanza di quattro millimetri. Quindi, preparare una miscela di trasfezione al plasma aggiungendo quattro microgrammi di ciascun plasmide a un volume totale di 50 microlitri di mezzo di elettroporazione.
Trasferire la miscela di trasfezione nella cuvetta, mescolare delicatamente sfiorando ed elettropolare con un impulso a 280 volt. Infine, trasferire 150 microlitri di fibroblasti elettroporati nella piastra di coltura cellulare di 10 centimetri preparata. Agitare la piastra tre volte in tutte le direzioni per distribuire le cellule e incubare durante la notte.
I fibroblasti sono presenti in un lungo fenotipo simile a un fuso, delle dimensioni da 150 a 300 nanometri. Dopo la riprogrammazione, si verificano colonie con piccole hiPSC di 50 micrometri che mostrano bordi distinti. Le hiPSCs si distinguono per un elevato rapporto nuclei/corpo e un aumento del tasso di proliferazione.
