Para começar, pegue um frasco de células de fibroblastos cultivadas, cubra uma placa de cultura de células de 10 centímetros adequada para cultura de hiPSC com quatro mililitros de solução de revestimento a frio por placa e incube a placa por uma hora a 37 graus Celsius. Em seguida, retirar os meios dos fibroblastos e lavá-los com quatro mililitros de PBS pré-aquecido por frasco. Aspirar o PBS e associar os fibroblastos com quatro mililitros de tripsina-EDTA por frasco incubando por cinco a sete minutos a 37 graus Celsius.
Monitorar o descolamento celular usando um microscópio de contraste de fase e, se necessário, bater contra a lateral do frasco para desprender as células da superfície plástica. Transfira as células destacadas para um tubo cônico de 50 mililitros e lave os frascos vazios com seis mililitros de meio fibroblasto pré-aquecido. Coletar e agrupar as células restantes no tubo cônico.
Em seguida, centrifugar as células a 200 g por cinco minutos a 20 graus Celsius. Aspirar o sobrenadante e ressuspender o pellet de células em três mililitros de PBS fresco pré-aquecido. Conte as células, transfira 1,5 vezes 10 para o sexto fibroblasto em um novo tubo cônico e encha com PBS até a marca de cinco milímetros.
Centrifugar o tubo a 200 g durante cinco minutos a 20 graus Celsius. Descarte o sobrenadante, ressuspenda o pellet celular em cinco mililitros de meio de eletroporação e centrifuja novamente. Enquanto isso, aspirar a solução de revestimento da placa de cultura celular revestida de 10 centímetros e adicionar sete mililitros de meio de fibroblastos pré-aquecido.
Ao final da centrifugação, descartar o sobrenadante e ressuspender o pellet celular em meio de eletroporação com concentração de 1,5 vezes 10 até a sexta célula em 250 microlitros. Transfira a suspensão de células de 250 microlitros para uma cubeta de eletroporação com uma distância de quatro milímetros de intervalo. Em seguida, prepare uma mistura de transfecção de plasma adicionando quatro microgramas de cada plasmídeo a um volume total de 50 microlitros de meio de eletroporação.
Transfira a mistura de transfecção para a cubeta, misture suavemente agitando e eletropore com um pulso a 280 volts. Finalmente, transferir 150 microlitros dos fibroblastos eletroporados para a placa de cultura de células de 10 centímetros preparada. Agite o prato três vezes em todas as direções para distribuir as células e incube durante a noite.
Os fibroblastos se apresentam em um fenótipo longo, fusiforme, de 150 a 300 nanômetros. Após a reprogramação, ocorrem colônias com pequenas hiPSCs de 50 micrômetros exibindo bordas distintas. As hiPSCs distinguem-se por uma alta relação núcleo/corpo e aumento da taxa de proliferação.
