Préparer les animaux à l’essai d’hérédité de l’ARNi en choisissant trois ou quatre adultes gravides de Caenorhabditis elegans sous les plaques NGM standard de 35 millimètres ensemencées avec la bactérie OP50-1. Après quatre jours, lavez les vers adultes gravides des plaques dans des tubes de 1,5 millilitre à l’aide de 800 microlitres de tampon M9 complété par Triton X-100. Répétez le lavage et tirez les vers dans un tube.
Après avoir centrifugé les vers à 1 000 g pendant 1,5 à deux minutes, aspirez le surnageant en laissant 100 microlitres sans perturber la pastille de vers. Dans chaque tube contenant la pastille de ver, ajoutez 650 microlitres d’eau distillée double. Pour isoler les embryons de C. elegans de la population collectée, ajoutez 250 microlitres de la solution de blanchiment fraîchement préparée et démarrez une minuterie.
Toutes les une à deux minutes, faites tourner les tubes à fond. Au bout de cinq minutes, surveillez la dégradation des vers adultes sous le stéréoscope. Continuez à vortex jusqu’à ce que les vers soient complètement dissous et qu’il ne reste que des embryons.
Cela prend généralement six à sept minutes. Centrifuger immédiatement les tubes à 1 000 G pendant 1,5 minute et aspirer le surnageant, en laissant environ 50 à 100 microlitres de surnageant avec les embryons. Ajouter un millilitre de tampon M9 complété par Triton X-100 aux embryons et au vortex.
Centrifugez la suspension et répétez le lavage deux fois. Après le lavage final, aspirez le surnageant et laissez environ 100 microlitres dans le tube. Vaporisez les tubes contenant des embryons isolés et pipetez deux microlitres de chaque tube deux fois sur une lame de verre étiquetée.
Comptez les embryons à l’aide d’un compteur de comptage pour estimer la concentration d’embryons par microlitre. Pour démarrer une croissance semi-synchronisée de la population, transférez environ 250 embryons sur chaque plaque d’ARNi NGM contenant la bactérie E. coli HT115 avec des vecteurs GFP ou ARNi témoins. Laisser le liquide absorber.
Incuber la génération de P zéro traitée avec de l’ARNi pendant quatre jours à 21 degrés Celsius.
