Preparar os animais para o ensaio de herança de RNAi escolhendo três ou quatro adultos de Caenorhabditis elegans grávidas sob as placas NGM padrão de 35 milímetros semeadas com bactérias OP50-1. Após quatro dias, lave os vermes adultos gravídicos das placas em tubos de 1,5 mililitro usando 800 microlitros de tampão M9 suplementado com Triton X-100. Repita a lavagem e puxe as minhocas para dentro de um tubo.
Depois de centrifugar os vermes a 1.000 G por 1,5 a dois minutos, aspirar o sobrenadante deixando 100 microlitros sem perturbar a pelota do minhoco. A cada tubo contendo o pellet de minhoca, adicione 650 microlitros de água bidestilada. Para isolar os embriões de C.elegans da população coletada, adicione 250 microlitros da solução de branqueamento recém-preparada e inicie um temporizador.
A cada um ou dois minutos, agite os tubos completamente. Após cinco minutos, monitore a degradação de vermes adultos sob o estereoscópio. Continue o vórtice até que os vermes estejam completamente dissolvidos e restem apenas embriões.
Isso geralmente leva de seis a sete minutos. Centrifugar imediatamente os tubos a 1.000 G por 1,5 minutos e aspirar o sobrenadante, deixando aproximadamente 50 a 100 microlitros do sobrenadante com os embriões. Adicionar um mililitro de tampão M9 suplementado com Triton X-100 aos embriões e vórtice.
Centrifugar a suspensão e repetir a lavagem duas vezes. Após a lavagem final, aspirar o sobrenadante e deixar aproximadamente 100 microlitros no tubo. Vórtice os tubos com embriões isolados e pipete dois microlitros de cada tubo duas vezes em uma lâmina de vidro marcada.
Conte os embriões usando um contador de contagem para estimar a concentração de embriões por microlitro. Para iniciar um crescimento populacional semi-sincronizado, transfira aproximadamente 250 embriões para cada placa RNAi NGM contendo bactérias E.coli HT115 com GFP ou vetores de RNAi controle. Deixe o líquido absorver.
Incubar a geração de P naught tratada com RNAi por quatro dias a 21 graus Celsius.
