Pour commencer le test à l’aide de l’agoniste des récepteurs plaquettaires, le peptide réticulé lié au collagène, ou CRPXL, diluer une quantité appropriée de stock de CRPXL dans le tampon de dosage à une concentration appropriée pour induire une agrégation plaquettaire maximale. La solution résultante sera dorénavant désignée sous le nom de norme. Ensuite, à partir des mêmes concentrations préparées les plus élevées, effectuez séquentiellement des dilutions en série deux fois pour l’étalon et le réactif d’essai.
Préparez une série de dilution de six à huit points pour l’essai et l’étalon dans le tampon d’essai qui génère des réponses d’agrégation de 100 % à 0 %Lorsque le plasma riche en plaquettes, ou PRP, est prêt à l’emploi, ajoutez une aliquote de PRP dans la cuvette d’agrégation de transmission de la lumière. Laissez les échantillons se réchauffer à 37 degrés Celsius dans les puits de rétention pendant quelques minutes. Une fois bien réchauffées, placez les cuvettes dans l’agrégateur pour commencer à enregistrer les traces.
En remuant constamment, ajoutez l’agoniste et enregistrez les données. Effectuer la collecte de données pour chaque dilution préparée de l’étalon et du réactif d’essai. Utilisez Graphpad Prism, ou tout autre logiciel approprié, pour vérifier que les courbes de concentration de l’étalon et de l’essai sont parallèles.
Entrez les données de manière à ce que l’axe des abscisses contienne les concentrations et que l’axe des ordonnées contienne les réponses indiquées soit par le pourcentage d’agrégation maximale, soit par l’aire sous la courbe. Ensuite, transformez les valeurs de concentration en logarithmes correspondants, ou concentration logarithmique. Sélectionnez la régression non linéaire à partir des fonctions d’analyse et utilisez l’équation pour la stimulation dose-réponse.
Sélectionnez l’agoniste logarithmique par rapport à la pente variable de réponse, et utilisez un ajustement à trois ou quatre voies, selon ce qui convient le mieux aux données. Déterminez si les courbes générées sont parallèles en cliquant sur l’onglet Comparer et en comparant les jeux de données. Assurez-vous que la pente et les asymptotes de l’essai, ainsi que les courbes standard, sont équivalentes.
Si ces conditions sont remplies, procédez au calcul de la puissance, ou CE50, à l’aide d’un ajustement de courbe de régression non linéaire. Calculez la puissance relative en divisant la valeur CE50 de l’étalon par celle de l’essai. Ajustez la concentration en masse par volume du réactif d’essai pour tenir compte de la différence de puissance par rapport à l’étalon.
Les traces d’agrégation plaquettaire pour l’étalon et le réactif test ont révélé la différence entre les deux réponses d’agrégation. À 0,075 microgramme par millilitre, il y avait une réponse claire à la norme, tandis que, pour le test, la concentration correspondante n’a montré aucune réponse. Les valeurs de la CE50 de l’étalon et du réactif d’essai ont été calculées en traçant la courbe de réponse à la concentration.
