Pour commencer, placez l’appareil gut-on-a-chip dans un four sec réglé à 60 degrés Celsius pendant 30 minutes pour éliminer l’humidité. Ensuite, à l’aide d’un générateur d’ozone UV, exposez l’appareil à un traitement UV et à l’ozone pendant 60 minutes. Fixez les tubes d’entrée, de dérivation et de sortie du microcanal supérieur à l’aide de pinces de liaison.
Plantez le tube d’entrée pour le microcanal inférieur, puis débranchez le tube de sortie pour le microcanal inférieur. Assurez-vous que le tube de dérivation du microcanal inférieur reste ouvert. À l’aide d’une micropipette P100, ajoutez 100 microlitres d’une solution de polyéthylénimine à 1 % à travers l’orifice de sortie du microcanal inférieur.
Rebranchez le tube de sortie du microcanal inférieur. Fixez le tube de dérivation d’entrée et de sortie du microcanal inférieur, suivi du tube d’entrée du microcanal supérieur à l’aide de clips de liant. Débranchez maintenant le tube de sortie du microcanal supérieur, en veillant à ce que le tube de dérivation reste ouvert.
Ensuite, ajoutez 100 microlitres d’une solution de polyéthylénimine à 1 % à travers le trou de sortie du microcanal supérieur. Remettez en place le tube de sortie du microcanal supérieur et laissez l’appareil incuber à température ambiante pendant 10 minutes. Répétez la procédure avec 0,1 % de glutaraldéhyde, puis avec de l’eau déminéralisée pour éliminer tout excès de solution d’activation de surface.
Séchez la puce pendant la nuit à 60 degrés Celsius dans un four. Le lendemain, sortez la puce du four et laissez-la refroidir dans une enceinte de sécurité biologique pendant 10 minutes. Fixez les tubes d’entrée, de dérivation et de sortie du microcanal supérieur, y compris le tube d’entrée du microcanal inférieur, à l’aide de pinces de liaison.
Débranchez le tube de sortie du microcanal inférieur. Ajoutez 20 microlitres de matrice extracellulaire, ou mélange ECM, à travers le trou de sortie du microcanal inférieur. Remettez ensuite en place le tube de sortie pour le micro-canal inférieur.
Fixez les tubes d’entrée, de dérivation et de sortie du microcanal inférieur, y compris le tube d’entrée du microcanal supérieur avec des clips de liant. Après avoir débranché le tube de sortie du microcanal supérieur, ajoutez 20 microlitres de mélange VCM à travers le trou de sortie du microcanal supérieur. Remettez en place le tube de sortie du microcanal supérieur, puis retirez le clip de liaison du tube de sortie du canal inférieur, en vous assurant que les deux tubes de sortie sont ouverts.
Placez la puce dans un incubateur à 37 degrés Celsius à 5 % de dioxyde de carbone pendant une heure.
