Fest to striper med 1,25 centimeter mikroporetape over toppen av krukken og legg lokket forsiktig på toppen. Forsegl gapet mellom lokket og krukken med 2,5 centimeter mikroporetape for å opprettholde steriliteten mens du tillater luftutveksling, og plasser krukken i et vekstkammer. Når plantene når ønsket alder, fjern mikroporebåndet og lokket fra krukken.
Aliquot 20 mikroliter vekstmedium på LB Agar Plate for sterilitetstesting. Bruk en 25 milliliter volumetrisk pipette, fjern så mye vekstmedium som mulig uten å skade røttene. Tilsett 50 ml oppsamlingsmedium langs krukkeveggen for å forhindre fukting av bladene.
Lukk krukken med lokket og inkuber i sterile forhold i to timer. Etter inkubering, fjern ønsket mengde oppsamlingsmedium i rør. Mål vekten av roten og skyt fra planten i en krukke for å normalisere roten ekssudater av plantevekt.
Arabidopsis thaliana planter så sunt ut over to forskjellige laboratorier. Glasskrukkesystemet viste seg å være tilpasningsdyktig for ulike plantearter og utviklingsstadier. Hvete hadde de høyeste rutevektene, etterfulgt av durra og tomat.
Mens durra hadde de høyeste rotvektene, etterfulgt av hvete og Medicago truncatula. Inokulering av Arabidopsis thaliana med kommensale bakterier endret ikke plantefenotypen, og inokulerte bakterier vedvarte i systemet i minst 12 dager med en liten økning i kolonidannende enheter.
