Per iniziare, installare camere da 0,5 millilitri nel respirometro O2K secondo le istruzioni del produttore. Accendere lo strumento e collegarlo al software in dotazione per l'acquisizione dei dati. Lavare le camere tre volte con acqua deionizzata.
Sostituire l'acqua con 0,54 millilitri di MiR05 e chiudere completamente i tappi. Dopo aver rimosso il tampone in eccesso, sollevare i tappi per consentire all'ossigeno ambiente di equilibrarsi con l'ossigeno della camera. Posizionare una colonna nel campo magnetico di un separatore di celle magnetiche e lavare la colonna con tre millilitri di RP5.
Risospendere la cellula mononucleata del sangue periferico appena isolata, o pellet PBMC, in 80 microlitri di RP5, insieme a 20 microlitri di microsfere anti CD14. Incubare le cellule per 15 minuti a quattro gradi Celsius. Diluire le celle con un millilitro di RP5 prima di caricare la sospensione sulla colonna.
Raccogliere le cellule non etichettate che fluiscono in un tubo conico da 15 millilitri etichettato come flusso attraverso uno"Lavare la colonna tre volte con tre millilitri di RP5 per raccogliere il flusso attraverso il liquido. Rimuovere con cautela la colonna dal campo magnetico e posizionarla su un nuovo tubo conico da 15 millilitri. Dopo aver aggiunto cinque millilitri di RP5, spingere lo stantuffo nella colonna per raccogliere il contenuto nella provetta.
Dopo aver centrifugato il flusso attraverso una provetta, ripetere i passaggi di isolamento utilizzando microsfere anti CD3 con le cellule per ottenere i linfociti T. Centrifugare le cellule contenenti cellule T CD3 positive e monociti CD14 positivi. Dopo aver scartato il surnatante, risospendere il pellet in un millilitro di RP5.
Utilizzando un emocitometro, determinare la concentrazione cellulare. Aggiungi 2,5 milioni di monociti e 5 milioni di cellule T in nuove provette da centrifuga. Pellettare le celle e rimetterle in sospensione in 20 microlitri di MiR05.
Installare i sensori fluorescenti e avviare un file sperimentale sull'unità respirometro O2K. Utilizzando i sensori fluorescenti a LED verde, con il filtro AMR impostato, regolare il guadagno del fluorimetro e l'intensità a 1000. Imposta il volume della camera su 0,5 millilitri e regola il layout per vedere sia il flusso di ossigeno che la fluorescenza TMRM.
Una volta che il flusso di ossigeno è stabile, eseguire la calibrazione dell'ossigeno nell'aria secondo il protocollo del produttore. Chiudere i tappi per sigillare la camera. Utilizzando una siringa di Hamilton, iniettare 2,5 microlitri di estere metilico di tetrametilrodamina 0,05 millimolari, o TMRM, quattro volte.
Tarare il sensore di flusso con un segnale fluorescente per ogni iniezione che rappresenta 0, 0,25, 0,5, 0,75 e 1,0 micromolari di TMRM. Iniettare 20 microlitri di MiR05 per eseguire l'esperimento in bianco. Dopo che il flusso di ossigeno si è stabilizzato, selezionare ed etichettare sia il flusso di ossigeno che il segnale TMRM come pre-cella.
Iniettare 20 microlitri della sospensione cellulare contenente le cellule e misurare per circa 10 minuti. Quindi selezionare ed etichettare sia il flusso di ossigeno che i segnali TMRM come cellule. Quindi ripetere i passaggi per ogni trattamento in sequenza ed etichettare il flusso di ossigeno e il segnale TMRM in modo appropriato.
Una volta che il flusso di ossigeno è stabile, iniettare un microlitro di antimicina A da 1,25 millimolari per inibire la respirazione mitocondriale. Per ogni esperimento in bianco, impostare la concentrazione TMRM pre-campione su uno per calcolare il rapporto di fondo. Calcola la successiva diminuzione proporzionale del TMRM e il rapporto medio di fondo da tutti gli esperimenti in bianco.
Quindi moltiplicare il TMRM pre-campione dell'esperimento campione con il rapporto di fondo medio per ogni iniezione di ciascun esperimento campione. Sottrarre il background dell'esperimento con i valori TMRM misurati del campione. Successivamente, sottrarre il potenziale di membrana mitocondriale corretto dal background FCCP da ogni iniezione.
Per normalizzare la diminuzione guidata dall'ADP, impostare il potenziale di membrana più alto e più basso rispettivamente come 100% e 0% e adattare i dati in un modello di regressione di adattamento non lineare. Studi comparativi sui cambiamenti indotti dall'ADP nelle cellule T e nei monociti di volontari sani hanno dimostrato che i monociti mostravano una maggiore perdita del potenziale di membrana mitocondriale rispetto alle cellule T. La concentrazione inibitoria semimassima di ADP sul potenziale di membrana mitocondriale era inferiore per i monociti rispetto alle cellule T.
Un tipico aumento dose-risposta dei tassi di consumo di ossigeno con ADP non è stato rilevato in nessuno dei due tipi di cellule.
