Con una jeringa Hamilton, inyecte 2,5 microlitros de 0,05 milimolares de tetrametilrodamina metil éster, o TMRM, cuatro veces.
Calibre el sensor de flujo con una señal fluorescente para cada inyección que represente 0, 0,25, 0,5, 0,75 y 1,0 micromolares de TMRM. Inyecte 20 microlitros de MiR05 para ejecutar el experimento en blanco. Después de que el flujo de oxígeno se estabilice, seleccione y etiquete tanto el flujo de oxígeno como la señal TMRM como pre-celda.
Inyecte 20 microlitros de la suspensión celular que contiene las células y mida durante unos 10 minutos. Para normalizar la disminución impulsada por el ADP, establezca el potencial de membrana más alto y más bajo en 100% y 0% respectivamente, y ajuste los datos en un modelo de regresión de ajuste no lineal.
