Weefsel stijfheid is een belangrijke determinant van cellulaire functie, en veranderingen in weefsel stijfheid worden vaak geassocieerd met fibrose, kanker en hart-en vaatziekten 1-11. Traditionele cel biologische benaderingen aan het bestuderen van cellulaire functie door culturen van cellen op een vaste ondergrond (plastic borden of glas dekglaasjes) die niet kan goed zijn voor het effect van een elastische ECM of de variaties in ECM stijfheid tussen de weefsels. Om het model in vivo weefsel compliance in vitro, wij en anderen gebruiken ECM-gecoate hydrogels. In ons laboratorium worden de hydrogelen op basis van polyacrylamide die het bereik van het weefsel naleving biologisch 12 gezien kan nabootsen. &quot;Reactieve&quot; dekglaasjes worden gegenereerd door incubatie met NaOH gevolgd door toevoeging van 3-APTMS. Glutaraldehyde wordt gebruikt om cross-link van de 3-APTMS en de polyacrylamide gel. Een oplossing van acrylamide (AC), bis-acrylamide (Bis-AC) en ammoniumpersulfaat wordt gebruikt voor de polymerisatie van de hydrogel. N-hydroxysuccinimide (NHS) is opgenomen in de AC-oplossing voor het verknopen ECM eiwit aan de hydrogel. Na polymerisatie van de hydrogel, is de gel oppervlak bedekt met een ECM-eiwit van de keuze, zoals fibronectine, vitronectine, collageen, etc. De stijfheid van een hydrogel kan worden bepaald door reologie of atomic force microscopie (AFM) en aangepast door het variëren van het percentage van de AC-en / of bis-AC in de oplossing 12. Op deze manier kan substraat stijfheid worden afgestemd op de stijfheid van de biologische weefsels, die ook kan worden gekwantificeerd aan de hand reologie of AFM. Cellen kunnen vervolgens worden ingedeeld op deze hydrogelen en gekweekt op basis van de vereiste experimentele omstandigheden. Beeldvorming van de cellen en hun herstel voor moleculaire analyse is eenvoudig. Voor dit artikel, definiëren we zachte ondergrond als die met elastische moduli (E) &lt;3000 Pascal en stijve ondergrond / weefsels als die met E&gt; 20.000 Pascal.
