Organotypische hippocampale slice cultuur is een in vitro methode om de mechanismen van de neuronale schade in de basisarchitectuur en samenstelling van de hippocampus relatief behouden blijft een te onderzoeken. Het organotypische cultuur systeem maakt het mogelijk voor het onderzoek van neuronale, astrocytaire en microgliale effecten, maar als een ex vivo voorbereiding, wordt niet ingegaan op effecten van de bloedstroom, of aanwerving van perifere inflammatoire cellen. Te dien einde, is deze cultuur methode vaak gebruikt om excitotoxisch en hypoxische schade aan piramidale neuronen van de hippocampus te onderzoeken, maar is ook gebruikt om de ontstekingsreactie te onderzoeken. Hierin beschrijven we de methoden voor het genereren van de hippocampus slice culturen van postnatale knaagdieren hersenen, het toedienen van giftige stimuli om neuronale letsel veroorzaken, en het testen en kwantificeren van hippocampale neuronale dood.
