Flera kritiska faktorer för en lyckad medulloblastom cell allografting som ger sekundära tumörbildning är följande: Först använder endast 50% Accutase för vävnadsdissociering och inte över-smälta den vävnad som långvarig enzym behandling leder till betydande minskning av cellernas livskraft. intracranial orthotopic allografting of medulloblastoma cells in immunocompromised mice

Medulloblastom är den vanligaste pediatrisk tumör i nervsystemet. En mängd djurstudier har fokuserat på cerebellär prekursorer granulat neuron (CGNPs) som cellen om ursprungsland för medulloblastom 1-4. Men de olika kliniska presentationer av medulloblastom subtyper hos människor (nodulär, Desmoplastiskt, klassisk och stor cell / anaplastisk), och det faktum att medulloblastom finns i en delmängd av mänskliga patienter utan ektopisk uttryck av CGNP markör 5, tyder på att cellulära och molekylära ursprunget medulloblastom är mer komplexa och långt ifrån fullständigt dechiffreras. Därför är det viktigt att avgöra om det finns ett alternativ medulloblastom tumörcellen om ursprungsland baserat på vilken celltyp specifika terapeutiska modaliteten kan utvecklas. För detta ändamål kommer intrakraniell orthotopic allografting av genetiskt märkta tumör celltyper följt av efterföljande analyser av sekundär tumörutveckling hos mottagare kunna bestämma den cellulära ursprunget av tumör-initiera celler. Här beskriver vi den experimentella protokoll för intrakraniell orthotopic allografting av medulloblastom celler från primära tumören, och detta förfarande kan även användas för att transplantera celler från etablerade cellinjer.
