Er zijn twee belangrijke stappen in het knaagdier WEC voor het succes. Ten eerste moet de dissectie procedure nauwkeurig niet te beschadigen embryo&#39;s, in het bijzonder de bloedvaten. Ten tweede moet de procedure worden zo snel mogelijk, omdat zuurstof en voedingsstoffen worden niet meer gevoed via de placenta na isolatie uit de baarmoeder. Hele embryo cultuur (WEC) techniek is ontwikkeld in 1950 is door de New en zijn collega&#39;s, en toegepast voor ontwikkelingsbiologie 1. Hoewel de ontwikkeling en groei van zoogdieren embryo&#39;s zijn sterk afhankelijk van de functie van de placenta, WEC-techniek stelt ons in staat om de cultuur muizen en ratten embryo&#39;s ex vivo conditie tijdens een beperkte periode die overeenkomt met midgestation etappes tijdens de embryonale dag (E) 6.5-E12.5 in de muis of E8.5-E14.5 in de rat 2, 3, 4. In de WEC, kunnen we direct richten gewenste gebieden van embryo&#39;s met behulp van fijne glazen capillairen, omdat embryo&#39;s kunnen worden gemanipuleerd onder de microscoop.
