patch clamp and perfusion techniques for studying ion channels expressed in xenopus oocytes

Le protocole présenté ici est conçu pour étudier l&#39;activation de la conductance grande tension et de Ca 2 +-K + activé (BK) canaux. Le protocole peut également être utilisé pour étudier la relation structure-fonction pour d&#39;autres canaux ioniques et les récepteurs des neurotransmetteurs 1. Canaux BK sont largement exprimés dans différents tissus et ont été impliqués dans de nombreuses fonctions physiologiques, y compris la réglementation de la contraction des muscles lisses, la fréquence d&#39;accord de cellules ciliées internes et la réglementation de libération des neurotransmetteurs 2-6. Canaux BK sont activés par dépolarisation de la membrane et par intracellulaire de Ca 2 + et Mg 2 + 6-9.
