Eines der wichtigsten Ziele in den Neurowissenschaften ist es, die molekularen Signale, die frühen Stadien der Synapsenbildung anweisen, zu verstehen. Als solches ist es unumgänglich geworden, um objektive Ansätze zu entwickeln, um Veränderungen der synaptischen Verbindungen zu quantifizieren. Ausgehend vom Beispiel Fixierung dieses Protokoll Details, wie man Synapse Anzahl sowohl in dissoziierten neuronalen Kultur und in Hirnschnitten mittels Immunzytochemie quantifizieren. Die Zahl dieser Kolokalisationen wird unter Verwendung eines Plug in Puncta Analyzer (geschrieben von Bary Wark, auf Anfrage, c.eroglu @ cellbio.duke.edu) unter dem ImageJ-Software-Plattform. Die Synapse-Assay in diesem Protokoll beschrieben, kann auf jeden Nervengewebe oder Kultur Vorbereitung, für die Sie selektiv prä-und postsynaptischen Markern angewendet werden. Diese Synapsen Assay ist ein wertvolles Werkzeug, das weit in das Studium der synaptischen Entwicklung genutzt werden kann.
