organotypic slice culture of gfp-expressing mouse embryos for real-time imaging of peripheral nerve outgrowth

Pour de nombreux usages, la culture de la souris ex vivo des embryons en tranches organotypiques est souhaitable. Par exemple, nous employons une lignée de souris transgéniques (tauGFP) dans lequel la version améliorée de la protéine fluorescente verte (EGFP) est exclusivement exprimé dans tous les neurones du système nerveux central en développement et périphérique 1, donnant la possibilité à la fois film de l&#39;innervation du la patte avant et de manipuler ce processus avec des techniques pharmacologiques et génétiques 2. Le paramètre le plus critique dans la culture réussie des cultures tranche telle est la méthode par laquelle les tranches sont préparées. Après de nombreux essais sur une variété de méthodes, nous avons constaté qu&#39;un vibratome est le meilleur dispositif possible de couper les embryons tels qu&#39;ils systématiquement lieu à une culture qui démontre la viabilité sur une période de plusieurs jours, et surtout, se développe dans un âge de manière spécifique. Pour la mi-gestation d&#39;embryons, ce qui inclut l&#39;excroissance normale des nerfs rachidiens de la moelle épinière et les ganglions de la racine dorsale de leurs objectifs dans la périphérie et la détermination adéquate des tissus squelettiques et musculaires. Dans ce travail, nous présentons une méthode pour le traitement des embryons entiers de jour embryonnaire (E) E10 à E12 dans 300 à 400 tranches micrométrique pour la culture dans une norme incubateur de culture tissulaire, ce qui peut être étudiée pour un maximum de deux jours après la préparation tranche. Critique pour le succès de cette approche est l&#39;utilisation d&#39;un vibratome de découper chaque embryon agarose embarqués. Elle est suivie par la culture des tranches sur les inserts de culture Millicell membrane placée sur un petit volume de milieu, résultant en une technique de culture de l&#39;interface. Une litière avec une moyenne de 7 embryons produit régulièrement au moins 14 tranches (2-3 tranches de la région forelimb par embryon), qui varie légèrement en raison de l&#39;âge des embryons ainsi que de l&#39;épaisseur des tranches. Environ 80% des tranches de culture montrent prolongement nerveux, qui peut être mesurée througout les 2 période de culture. Les résultats représentatifs en utilisant la lignée de souris tauGFP sont démontrés.
