Este método se basa en el método descrito por primera vez por Stoppini et al. 11 y ofrece una manera rápida a las rebanadas de hipocampo cultura. organotypic hippocampal slice cultures

El hipocampo, un componente del sistema límbico, juega un papel importante en la memoria a largo plazo y la navegación espacial 1. Las neuronas del hipocampo puede modificar la fuerza de sus conexiones después de breves períodos de fuerte activación. Este fenómeno, conocido como potenciación a largo plazo (LTP) puede durar horas o días y se ha convertido en el mecanismo mejor candidato para el aprendizaje y la memoria 2. Además, la anatomía bien definida y la conectividad del hipocampo 3 se ha convertido en un sistema modelo clásico para estudiar la transmisión sináptica y la plasticidad sináptica 4. A medida que nuestra comprensión de la fisiología de las sinapsis del hipocampo creció y los jugadores molecular se identificó, la necesidad de manipular las proteínas sinápticas se convirtió en un imperativo. Organotípicos culturas del hipocampo ofrecen la posibilidad de la manipulación genética fácil y precisa la intervención farmacológica, sino mantener la organización sináptica que es fundamental para comprender la función de la sinapsis en un contexto más natural que los métodos de rutina de la cultura neuronas disociadas. Aquí presentamos un método para preparar y cultura rodajas de hipocampo que se pueden adaptar fácilmente a otras regiones del cerebro. Este método permite un fácil acceso a los cortes para la manipulación genética por métodos diferentes como la infección viral o biolística 5,6 7. Además, los cortes pueden ser fácilmente recuperados para los ensayos bioquímicos 8, o transferirse a microscopios para imágenes 9 o 10 experimentos electrofisiológicos.

Se describe un método para preparar organotípicos rodajas de hipocampo que se pueden adaptar fácilmente a otras regiones del cerebro. Cortes de cerebro se ponen en membranas porosas y medios de cultivo se le permite formar una interfase.
