Biofilmer odlas i olika reaktorer kommer ofta har olika egenskaper och varje reaktor har olika användningsområden. I detta arbete beskriver vi att använda två biofilm reaktorer: dropp flöde biofilm reaktor och en roterande skiva reaktor. De flesta mikrober i naturen är tänkt att finnas som underlag-associerad samhällen i biofilmer. 1 Bakteriella biofilmer är inneslutna i en matris och knytas till en yta. 2 biofilm bildning och utveckling ofta studeras i laboratorium med parti system som mikrotiterplattor eller flöde system, såsom flow-celler. Dessa metoder är användbara för screening mutant och kemiska bibliotek (mikrotiterplattor) 3 eller växande biofilmer för visualisering (flöde celler) 4. Här presenterar vi detaljerade protokoll för växande Staphylococcus aureus i ytterligare två typer av biofilmer flödessystem: dropp flödet biofilmen reaktorn och roterande skiva biofilmen reaktor. Drip flöde biofilm reaktorer är konstruerade för att studera biofilmer odlas under låg skjuvning villkor. 5 droppa flödet Reaktorn består av fyra parallella testa kanaler, var och kan hålla en standard glas objektglas storlek kupong, eller en längd av kateter eller snåla. Droppet flödet reaktorn är idealisk för microsensor övervakning, allmän biofilm studier, biofilm cryosectioning prover, hög produktion av biomassa, medicinska utvärderingar material och inneboende medicintekniskt testning. 6,7,8,9 Den roterande skiva Reaktorn består av en teflon-skiva som innehåller urtag för flyttbara kuponger. 10 Den avtagbara kuponger som kan göras från någon bearbetbara material. I botten av den roterande skivan innehåller en stavmagnet att låta disken rotera för att skapa vätskeytan klippa över ytan-flush kuponger. Hela skiva som innehåller 18 kuponger placeras i en 1000 ml glasflaska sida armen reaktortanken. En flytande tillväxt media cirkulerar genom fartyget medan skivan roteras med en magnetomrörare. Kupongerna tas bort från reaktortanken och sedan skrapade samla biofilmen prov för vidare studier eller mikroskopi avbildning. Roterande skiva reaktorer är konstruerade för Laboratoriebedömning av biocid effekt, biofilm borttagning, och prestanda för antifoulingsystem material. 9,11,12,13
