Neurogenese in de postnatale hersenen hangt af van het onderhoud van drie biologische gebeurtenissen: proliferatie van stamcellen, migratie van neuroblasts, evenals differentiatie en integratie van nieuwe neuronen in bestaande neurale circuits. Voor de postnatale neurogenese in de olfactorische bollen, zijn deze gebeurtenissen gescheiden binnen drie anatomisch verschillende domeinen: proliferatie grotendeels komt voor in de subependymal zone (SEZ) van de laterale ventrikels, migreren neuroblasts dwars door de rostrale migratie stroom (RMS), en nieuwe neuronen differentiëren en te integreren in de olfactorische lampen (OB). De drie domeinen dienen als ideaal platform om de cellulaire, moleculaire en fysiologische mechanismen die duidelijk regelen van elk van de biologische gebeurtenissen te bestuderen. Dit artikel beschrijft een organotypische slice assay geoptimaliseerd voor postnatale hersenweefsel, waarin de extracellulaire voorwaarden nauw aan bij de na te bootsen in vivo omgeving te migreren neuroblasts. We tonen aan dat onze test voorziet in een uniforme, georiënteerd, en snelle beweging van neuroblasts binnen de RMS. Deze test zal zijn uitermate geschikt voor de studie van de cel-autonome en niet-zelfstandige regulering van de neuronale migratie door gebruik te maken van cross-transplantatie benaderingen uit de muizen op verschillende genetische achtergronden.
