Протокол состоит из трех частей: изоляции дыхательных путей эпителиальные клетки мыши трахеи, культивирования этих клеток на воздух-жидкость (ALI) как полностью дифференцированные клетки эпителия, а также доставку калиброванный основной CS этих клеток в культуре. Система АЛИ культуры позволяет культуры эпителия дыхательных в условиях, которые больше напоминают их физиологические настройки по сравнению с обычными системами культуральной жидкости. Изучение молекулярных и легких клеточных реакций на воздействие CS является критически важным компонентом понимания воздействия загрязнения окружающей среды воздуха на здоровье человека.
