Pulmonell epitelceller kan isoleras från luftvägarna hos möss och odlade på luft-vätska gränssnitt (ALI) som en modell för differentierade respiratoriska epitelet. Ett protokoll beskrivs för att isolera och utsätta dessa celler att integrera cigarettrök (CS), för att studera epitelceller svar på CS exponering. Protokollet består av tre delar: isolering av luftvägarna epitelceller från mus luftstrupen, den odling av dessa celler vid luft-vätska gränssnitt (ALI) som fullt differentierade epitelceller, och leverans av kalibrerade mainstream CS till dessa celler i kultur. ALI kulturen systemet tillåter kulturen i andningsorganen epitel under förhållanden som mer liknar deras fysiologiska inställning än det vanliga flytande kultur. Studier av molekylära och lungor cellulära svaren på CS exponeringen är en kritisk komponent för att förstå effekterna av miljön luftföroreningar på människors hälsa.
